黑果枸杞花青素对H9c2大鼠心肌细胞抗氧化能力、炎性因子及能量代谢作用的影响 被引量:2

摘要 试验旨在探究青海柴达木黑果枸杞花青素(AC)对H9c2大鼠心肌细胞氧化应激损伤的影响,选用H9c2大鼠心肌细胞为研究对象,设立常氧组、常氧+AC组、低氧组和低氧+AC组,利用ELISA法检测各组细胞中过氧化物酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)、血红素加氧酶-1(HO-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和白细胞介素-10(IL-10)以及能量代谢相关因子Na^(+)-K^(+)-ATP酶、Ca^(2+)-ATP酶的变化。结果显示,与常氧组相比,常氧+AC组大鼠心肌细胞MDA、TNF-α、IL-6的含量均显著降低(P<0.05),CAT、HO-1、Na^(+)-K^(+)-ATP酶、Ca^(2+)-ATP酶活性显著升高(P<0.05),GSH-Px、SOD活性和IL-10含量均极显著升高(P<0.01);低氧组大鼠心肌细胞GSH-Px、HO-1、Na^(+)-K^(+)-ATP酶活性显著降低(P<0.05),CAT、SOD、Ca^(2+)-ATP酶活性和IL-10含量均极显著降低(P<0.01),MDA、TNF-α、IL-6含量均极显著升高(P<0.01)。与低氧组相比,低氧+AC组大鼠心肌细胞MDA、TNF-α、IL-6含量均极显著降低(P<0.01),CAT、GSH-Px、SOD、HO-1、Na^(+)-K^(+)-ATP酶、Ca^(2+)-ATP酶活性和IL-10含量均极显著升高(P<0.01)。研究表明,AC能够通过减轻H9c2大鼠心肌细胞氧化应激损伤,增强低氧下心肌细胞的能量代谢,抑制低氧诱导的炎症发生,进而缓解H9c2大鼠心肌细胞的低氧损伤。 The purpose of the experiment was to investigate the effect of Qinghai Qaidam AC on oxidative stress injury of H9c2 rat cardiomyocytes.The cardiomyocytes of H9c2 rats were selected as the research objects,and normal oxygen group,normal oxygen+AC group,hypoxia group and hypoxia+AC group were set up.The changes of CAT,GSH-Px,SOD,MDA,HO-1,TNF-α,IL-6 and IL-10,Na^(+)-K^(+)-ATPase and Ca^(2+)-ATPase were detected by ELISA.The results showed that compared with normal-oxygen group,the content of MDA,TNF-αand IL-6 in normal-oxygen+AC group were significantly decreased(P<0.05),and the activitiy of CAT,HO-1,Na^(+)-K^(+)-ATPase and Ca^(2+)-ATPase were significantly increased(P<0.05).GSH-Px,SOD activity and IL-10 content were extremely increased(P<0.01).In hypoxia group,the activity of GSH-Px,HO-1 and Na^(+)-K^(+)-ATPase were significantly decreased(P<0.05),the activity of CAT,SOD and Ca^(2+)-ATPase and the content of IL-10 were extremely decreased(P<0.01),and the content of MDA,TNF-αand IL-6 were extremely increased(P<0.01).Compared with hypoxia group,the content of MDA,TNF-αand IL-6 in hypoxia+AC group were extremely decreased(P<0.01),while the activity of CAT,GSH-Px,SOD,HO-1,Na^(+)-K^(+)-ATPase,Ca^(2+)-ATPase and the content of IL-10 were extremely increased(P<0.01).The study indicates that AC can alleviate hypoxic injury of H9c2 rat cardiomyocytes by alleviating oxidative stress injury,enhancing energy metabolism of cardiomyocytes under hypoxia,inhibiting hypoxicinduced inflammation,and thus alleviating hypoxic injury of H9c2 rat cardiomyocytes.
