摘要
目的 优化五味子中总木脂素的提取工艺,并对其主要成分进行含量测定。方法 以提取时间、料液比、微波功率和超声波功率为单因素,研究各因素对五味子总木脂素得率的影响;在此基础上,按L9(34)设计正交试验,对超声波微波协同辅助提取工艺进行优化;采用超高效液相色谱(ultra-high performance liquid chromatography,UPLC)法对主要成分进行含量测定。结果 五味子总木脂素最优提取工艺为:提取溶剂为80%的乙醇,料液比为1∶20,提取温度为60℃,微波功率为150 W,超声波功率为200 W,提取时间为30 min。在此条件下所得五味子总木脂素的含量和得率分别为48.31%和8.55%,五味子醇甲、五味子醇乙、五味子酯甲、五味子酯乙、五味子甲素、五味子乙素和五味子丙素7个化学成分含量分别为4.22%、1.04%、0.28%、0.88%、1.60%、1.71%和0.05%。结论 该方法准确性及重现性好,提取时间短,效率高,操作简便,可为后续五味子总木脂素功能性及其机制研究提供参考。
Objective:To optimize the extraction process of total lignans from Schisandra chinensis(Turcz.)Baill.fruits and determine the content of its main components.Methods:The extraction time,material-to-liquid ratio,microwave power and ultrasonic power were used as single factors to study the influence of each factor on the yield of total lignans.On the basis of single factor experiment,orthogonal experiment was designed according to L9(34)to optimize the extraction process assisted by ultrasonic-microwave synergistic extraction.The UPLC method was established to determine the main components.Results:The extraction process of total lignans from Schisandra chinensis was determined as follows:ethanol concentration 80%,material/liquid ratio 1∶20,extraction temperature 60℃,microwave power 150 W,ultrasonic power of 200 W and extraction time 30 min.Under these conditions,the content and yield of total lignans were 48.31%and 8.55%,respectively.The contents of Schisandrol A,Schisandrol B,Schisantherin A,Schisantherin B,Deoxyschisandrin,Schisandrin B,and Schisandrin C were 4.22%,1.04%,0.28%,0.88%,1.6%,1.71%and 0.05%,respectively.Conclusion:This study can provide a material basis for the study on the function and mechanism of total lignans in Schisandra chinensis.
