双波长HPLC法测定藤黄健骨片中地黄苷D、水晶兰苷的含量

Determination of Dihuang glucoside D and crystal blue glucoside in Tenghuang Jiangu Tablets by Dual Wavelength HPLC

目的为完善藤黄健骨片质量标准,建立双波长HPLC法同时测定藤黄健骨片中地黄苷D、水晶兰苷的含量.方法采用高效液相色谱法,色谱柱为SVEA opal C18柱(4.6mm×250mm,5μm),流动相为甲醇-0.1%磷酸(5:95),检测波长为203nm(地黄苷D)、235nm(水晶兰苷),柱温为30℃,流速为1.0mL·min^(-1),进样量均为10μl.结果地黄苷D、水晶兰苷分别在0.01502-0.4506μg(R^(2)=1.0000)、0.0108-0.3240μg(R^(2)=1.0000)范围内线性关系良好,精密度、重复性、稳定性良好,平均回收率分别为98.5%(RSD=0.62%,n=6)、98.5%(RSD=1.17%,n=6).结论该分析方法科学可行,专属性、重复性、稳定性均符合要求,可为藤黄健骨片中熟地黄、鹿衔草的质量控制提供参考.

Objective To improve the quality standard of Tenghuang Jiangu Tablets,a dual wavelength HPLC method was established for the simultaneous determination of Dihuang glucoside D and crystal blue glucoside in Tenghuang Jiangu Tablets.Methods Using HPLC,the chromatographic column is SVEA opal C18 column(4.6mm×250mm,5μm),the mobile phase is methanol-0.1%phosphoric acid(5:95),the detection wavelength is 203nm(Dihuang glucoside D),235nm(crystal blue glucoside),the column temperature is 30℃,the flow rate is 1.OmL·min^(-1),and the sample injection volume is 10μl.Results The linear ranges of Dihuang glucoside D and crystal blue glucoside are 0.01502-0.4506μg(R^(2)=1.0000),0.0108-0.3240μg(R^(2)=1.0000)respectively,good precision,repeatability and stability,the average recoveries were 98.5%(RSD=0.62%,n=6)and 98.5%(RSD=1.17%,n=6),respectively.Conclusion The analytical method is scientific and feasible,and its specificity,repeatability and stability meet the requirements.It can provide a reference for the quality control of Radix Rehmanniae Preparata and Herba Cervi in Tenghuang Jiangu Tablets.
