基于生物信息学分析2型糖尿病关键基因及免疫细胞浸润情况

摘要 目的应用生物信息学的方法筛选2型糖尿病(type 2 diabetes mellitus,T2DM)患者的关键基因及免疫细胞浸润情况。方法从GEO数据库下载基因表达数据集,利用多种算法分析差异表达基因(differentially ex⁃pressed genes,DEGs),进行功能富集,构建蛋白互作网络,鉴定关键基因,挖掘免疫细胞浸润情况等。结果在T2DM中共发现137个DEGs,筛选出COL1A1、COL1A2、COL3A1、COL18A1、COL7A1、PCOLCE2、TGFB3、THBS1、HLA-DRB1、HLA-DRB共510个关键基因。M1巨噬细胞、M2巨噬细胞、滤泡辅助性T细胞(Tfh)和中性粒细胞的比例明显高于对照组织,差异有统计学意义(P<0.05)。结论筛选出的10个关键基因和4种免疫细胞可能参与了T2DM的发生发展。 Objective To screen key genes and immune cell infiltration in patients with type 2 diabetes mellitus(T2DM)by applying a bioinformatics approach.Methods Gene expression datasets from GEO database were downloaded,differentially expressed genes(DEGs)were analyzed using various algorithms,functional enrichment was performed,protein interaction networks were constructed,key genes,and mined for immune cell infiltration were identified.Results A total of 137 DEGs were identified in T2DM,and 10 key genes were screened,including COL1A1,COL1A2,COL3A1,COL18A1,COL7A1,PCOLCE2,TGFB3,THBS1,HLA-DRB1,HLA-DRB5,etc.M1 macrophages,M2 macrophages,follicular helper T cells(Tfh)and neutrophils were significantly higher than those in control tissues,and the differences were statistically significant(P<0.05).Conclusion The 10 key genes and 4 immune cells screened may be involved in the development of T2DM.
