新鲜黄独中黄独素B的含量测定

摘要 研究了高效液相色谱法测定新鲜黄独的块茎、叶子和地上茎中黄独素B的含量。采用Ultimate XB-C_(18)柱(250×4.6 mmID,5μm),流动相为乙腈,检测波长220 nm,流速1.0 mL/min,柱温25℃。黄独素B在10.6~53.0μg/mL质量浓度范围内线性关系良好,回归方程为y=21.392x+1146.7,相关系数R为0.9992。经验证重复性、稳定性及精密度试验相对标准偏差RSD均符合要求,样品加标回收率试验RSD为1.4%,在允许范围内,故本方法适合新鲜黄独中黄独素B含量的测定。新鲜黄独块茎、叶子及地上茎中黄独素B的含量在本次测定中分别为0.122%,0.060%及0.012%。 We studied the determination of content of diosbulbin B in the tuber,leaf and stem of fresh dioscorea bulbifera L.by the method of HPLC.The Ultimate XB-C_(18)(250×4.6 mmID,5μm)column was used.The mobile phases is acetonitrile,the detection wavelength was 220 nm,the flow rate was 1.0 mL/min and the column temperature was 25℃.The linear concentration range of diosbulbin B was at 10.6~53.0μg/mL,the regression equation is y=21.392x+1146.7 and correlation coefficient R is 0.9992.The relative standard deviation(RSD)of repeatability,stability and precision test was in line with the requirements,and the RSD of the standard recovery test was 1.4%.Therefore,this method was suitable for the determination of content of Diosbulbin B in resh dioscorea bulbifera L.And the contents of the tuber,leaf and stem in fresh dioscorea bulbifera L.are 0.122%,0.060%and 0.012%respectively.
