摘要
利用生物信息学的方法在莱茵衣藻(Chlamydomonas reinhardtii)的全基因组中,鉴定出8个碳酸酐酶(Carbonic Anhydrase,CA)家族基因,分别在理化性质、同源性、保守基序和染色体定位等方面分析,并对莱茵衣藻培养过程中添加碳酸氢盐实验和相关基因的表达进行分析。结果显示:8个莱茵衣藻CA蛋白序列长度为264~399 aa,分子量为33.46~42.17 kDa,理论等电点pI介于5.7~9.05之间;莱茵衣藻CA基因家族分为3个亚族;莱茵衣藻的8个CA基因均含有特定保守基序;染色体定位发现基因Cr1和Cr2在4号染色体,Cr4和Cr5在5号染色体,Cr3和Cr6在9号染色体,Cr7和Cr8分别在12和13号染色体上;添加碳酸氢盐的实验和相对表达分析表明CA基因在利用无机碳发挥着重要的作用。
Bioinformatics analysis of the carbonic anhydrase(CA)protein family in chlamydomonas reinhardtii was conducted by using bioinformatics methods.Eight CA family genes were identified in the whole genome of chlamydomonas reinhardtii,and their physical and chemical properties,homology,conserved motifs,and chromosome locations were analyzed.The expression of related genes was analyzed during the cultivation process of chlamydomonas reinhardtii under the addition of bicarbonate.The results show that the length of the eight chlamydomonas reinhardtii CA protein sequences is between 264~399 amino acids,with molecular weights between 33.46~42.17 kDa,and theoretical isoelectric points(pI)ranging from 5.7 to 9.05.The chlamydomonas reinhardtii CA gene family is divided into three subfamilies,and all eight CA genes contained specific conserved motifs.Chromosome location analysis show that genes Cr1 and Cr2 are on chromosome 4,Cr4 and Cr5 are on chromosome 5,Cr3 and Cr6 are on chromosome 9,and Cr7 and Cr8 are on chromosomes 12 and 13,respectively.Experimental results and relative expression analysis of CA genes show that they play an important role in the utilization of inorganic carbon under the addition of bicarbonate during the cultivation process of chlamydomonas reinhardtii.
