基于网络药理学探讨百部-苦参协同抗菌作用机制 被引量:3

Action mechanism of synergistic antibacterial effect of Stemona japonica-Radix Sophorae Flavescentis based on network pharmacology

摘要 目的 基于网络药理学探讨百部-苦参协同抗菌作用机制。方法 通过TCMSP数据库分析百部-苦参主要活性成分,与Genecard数据库中的抗菌基因对比,筛选后将核心基因输入David数据库,进行基因本体(GO)生物学过程分析及基因组百科全书(KEGG)富集分析,运用Cytoscape3.7.1软件、String数据库等协助分析绘图。结果 研究获得百部28个、苦参25个抗菌关键活性成分,35个抗菌核心靶点。GO及KEGG分析发现,百部-苦参主要通过芒柄花黄素(Formononetin)等活性成分,作用于环加氧酶2(PTGS2)、环加氧酶1(PTGS1)、c-AMP依赖蛋白激酶A(PRKACA)、Jun原癌基因(JUN)、雌激素受体α(ESR1)等靶点,调节癌症通路、P53信号通路、T细胞受体信号通路及肿瘤坏死因子(TNF)信号通路。结论 百部-苦参协同抗菌体现了多成分、多靶点、多途径的特点,为进一步开展百部、苦参抗菌作用药效物质基础和作用机制研究提供了新的思路和方法。 Objective To explore the mechanism of synergistic antibacterial effect of Stemona japonica-Radix Sophorae Flavescentis based on network pharmacology.Methods The study analyzed the main active ingredients of Stemona japonica-Radix Sophorae Flavescentis through TCMSP database,compared with antimicrobial genes in Gen-ecard database,and after screening,the core genes were entered into David6.8 database for gene ontology(CO)bi-ological process analysis and KEGG enrichment analysis,acquiring the plots with the assistance of Cytoscape 3.7.1 software and String database.Results 28 antibacterial key active ingredients in Stemona japonica and 22 in Radix Sophorae Flavescentis were obtained in the study,along with 32 antibacterial core targets.Analysis of Go and KEGG revealed that by active ingredients such as Formononetin,Baibu-Kushen mainly acts oncyclooxygenase 2(PTGS2),cyclooxygenase 1(PTGS1),c-AMP-dependent protein kinase A(PRKACA),Jun proto oncogene(JUN),estrogen receptorα(ESR1),and other targets,regulating cancer pathways,p53 signaling,T cell receptor signaling,and tumor necrosis factor(TNF)signaling.Conclusion The synergistic antibacterial mechanism of Stemona japonica-Radix Sophorae Flavescentis features with multi-component,multi-target and multi pathway,which will provide a new idea and method for further research on the mechanism of the pharmacodynamics of the antibacterial effect of Stemona ja-ponica-Radix Sophorae Flavescentis.
