聚合7^(OE)亚基1B/1R易位系材料的创制及其品质研究 被引量:1

摘要 为提高小麦1B/1R易位系的加工品质,本研究选用高分子量麦谷蛋白亚基组成为1/7+9/2+12的小麦1B/1R易位系品种科农199为母本,与高分子量麦谷蛋白亚基组成为1/7^(OE)+8^(*)/5+10的强筋春小麦品种津强6号杂交,利用分子标记在F4代株系中筛选到一个7^(OE)亚基基因与黑麦碱基因聚合在同一条染色体上的1B/1R易位系材料。PCR结果显示,该聚合材料和非聚合材料在Glu-A1和Glu-D1位点没有差异。在温室种植条件下,对这些聚合材料和非聚合材料及其亲本进行麦谷蛋白溶胀指数(SIG)测定,结果表明,与1B/1R易位系亲本科农199比较,聚合5+10优质亚基后SIG值显著提高,聚合7^(OE)优质亚基后,SIG值进一步显著提高,部分聚合材料达到了强筋亲本津强6号的水平。将聚合材料和非聚合材料及其亲本种植于大田,发现聚合5+10和7^(OE)优质亚基株系的乳酸-SDS溶剂保持力(LA-SDS SRC)和SDS沉降值均显著高于只聚合5+10优质亚基的株系,但未达到强筋亲本津强6号的水平。综合来看,聚合了7^(OE)亚基的1B/1R易位系材料的加工品质显著提升。本研究为改良我国众多1B/1R易位系的加工品质提供了一项新的育种策略。 In order to improve the processing quality of wheat 1B/1R translocation lines,a cross was made between a wheat 1B/1R translocation line variety Kenong 199 with the high molecular weight glutenin subunit(HMW-GS)composition of 1/7+9/2+12 and a strong gluten spring wheat variety Jinqiang 6 with the HMW-GS composition of 1/7^(OE)+8^(*)/5+10.From an F_(4)generation line planted in a greenhouse,we screened some plants carrying the 7^(OE)subunit and the secalin genes pyramided on the same chromosome with molecular markers,without any polymorphism at Glu-A1 or Glu-D1 locus.Some pyramid and non-pyramid plants and their parents were selected for SIG determination,the results showed that compared with the parent Kenong 199,the SIG values of the 1B/1R translocation lines pyramiding 5+10 high-quality subunits were significantly increased.When 5+10 and 7^(OE)high-quality subunits were simultaneously pyramided into the 1B/1R translocation lines,the SIG values of partial pyramid plants increased further and reached the level of the strong gluten parent Jinqiang 6.The next generation was planted in the field,and the lines that pyramided 5+10 and 7^(OE)high-quality subunits had significantly higher lactic acid-SDS solvent retention capacity(LA-SDS SRC)and SDS sedimentation values than those only pyramiding 5+10 high-quality subunits,yet did not reach the level of the strong gluten parent.Taken together,the processing quality of the 1B/1R translocation lines carrying with the 7^(OE)subunit was significantly improved.This study provides a new breeding strategy for improving the processing quality of the widespread 1B/1R translocation lines in China.
