呼吸机联合中药茶包治疗OSAHS的疗效及其网络药理学机制探讨

Efficacy of ventilator combined with Chinese medicine tea bags in the treatment of OSAHS and its network pharmacological mechanism

目的通过回顾性研究在网络药理学的基础上探讨呼吸机联合中药茶包治疗OSAHS的疗效及机制。方法对2021年5月~2022年6月在安徽中医药大学第一附属医院门诊明确诊断为OSAHS的患者资料进行整理分析,筛选出符合条件的病例。并通过比较CTD及Gene Cards数据库下载OSAHS的相关基因数据;应用BATMAN-TCM数据库对中药茶包的活性成分及作用靶点进行预测分析,进而使用String构建共同靶点网络,对交集共同靶点基因用R 3.6.3软件进行富集分析,以预测中药茶包辅助治疗OSAHS的可能作用机制。结果(1)临床疗效:①治疗后鼾声强度下降的患者有25人,有效率为96.15%;②治疗前后嗜睡人数为(26/4);③治疗前后焦虑抑郁状态人数为(26/2);④治疗后无咽部不适、口干情况;(2)网络药理学:找到与OSAHS的相关靶点有1194个,在BATMAN-TCM中筛选出与养阴润喉茶相关的基因靶点299个,与喉症泻火茶相关的基因靶点432个,交集后,与养阴润喉茶有53个相关靶点,与喉症泻火茶有71个相关靶点。在KEGG和GO富集结果中,养阴润喉茶的显著治疗靶点在“神经元细胞体”和“流体剪切应力与动脉粥样硬化”,喉症泻火茶的显著治疗靶点在“神经递质受体活性”和“神经活性配体-受体相互作用”。结论中药茶包在OSAHS的治疗中可发挥显著的疗效。基于网络药理学,其对OSAHS疾病相关的多个靶点和多条信号通路的调控是提高OSAHS治疗有效率的重要作用机制。

Objective To investigate the efficacy mechanism of ventilator combined with Chinese herbal tea bags in the treatment of OSAHS through retrospective research on the basis of network pharmacology.Methods From May 2021 to June 2022,the data of patients who were clearly diagnosed as OSAHS in the outpatient clinic of the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine were sorted out and analyzed,and eligible cases were screened out.And download OSAHS related Gene data by comparing CTD and Gene Cards database,BATMAN-TCM database was used to predict and analyze the active ingredients and targets of Chinese medicine tea bags,and then String was used to construct a common target network,and R 3.6.3 software was used to enrich the genes of intersecting common targets,so as to predict the possible mechanism of action of Chinese medicine tea bags in the adjuvant treatment of OSAHS.Results(1)Clinical efficacy:①There were 25 patients with a decrease in snoring intensity after treatment,and the effective rate was 96.15%;②The number of sleepy people before and after treatment was(26/4);③The number of people with anxiety and depression before and after treatment was(26/2);④There was no pharyngeal discomfort and dry mouth after treatment.(2)Network pharmacology:1194 targets related to OSAHS were found,299 gene targets related to Yangyin throat tea and 432 gene targets related to laryngeal laxative tea were screened in BATMAN-TCM,and after intersection,there were 53 related targets with yin throat tea and 71 related targets with laryngeal laxative tea.In the results of KEGG and GO enrichment,the significant therapeutic targets of Yangyin Throat Tea were in“Neuronal Cell Body”and“Fluid Shear Stress and Atherosclerosis”,and the significant therapeutic targets of Laryngeal Epsomal Tea were in“Neurotransmitter Receptor Activity”and“Neuroactive Liand-Receptor Interaction”.Conclusions Chinese herbal tea bags can play a significant role in the treatment of OSAHS.Based on network pharmacology,its regulation of multiple targets and multiple signaling pathways related to OSAHS diseases is an important mechanism to improve the efficiency of OSAHS treatment.
