老年男性沃辛瘤Th淋巴细胞亚群及细胞因子检测的实验研究

Experimental study on Th lymphocyte subsets and cytokine detection in elderly male Warthin’s tumor

目的探讨沃辛瘤Th淋巴细胞亚群分布及细胞因子表达特点。方法收集32例沃辛瘤手术标本。采用免疫组化方法,分别应用抗T-bet、Gata3、ROR-γt、Foxp3抗体检测24例沃辛瘤标本中Th1、Th2、Th17、Treg淋巴细胞亚群。应用抗体芯片检测8例沃辛瘤细胞因子表达情况。结果Th1、Th2、Th17及Foxp^(3+)Treg阳性细胞占比(x±s,%)为2.48±2.129、0.70±1.101、0.97±1.002及6.26±3.567,Foxp^(3+)Treg细胞数量明显高于其他淋巴细胞亚群。沃辛瘤中趋化因子、淋巴归巢分子、淋巴滤泡形成相关因子表达较高,并且存在TNF-α、IL-1β及IL-6等重要衰老相关分泌表型,R语言分析40个细胞因子彼此间的相关性发现,这些衰老相关分泌表型因子与IL-8、IL-10、IL-12、GM-CSF、IL-11、IL-15及IL-6sR等炎性因子表达呈非常显著正相关性。结论沃辛瘤好发于老年患者,淋巴细胞亚群及细胞因子分布特点可能与其淋巴间质增龄性变化有关,据此推测淋巴间质增龄性变化在促进淋巴间质发展中可能起重要作用。

Objective To investigate the Th lymphocyte subsets distribution and cytokine expression characteristics in warthin’s tumor.Methods 32 surgical specimens of parotid Warthin's tumor were collected.The lymphocyte subsets of Th1,Th2,Th17 and Treg in 24 Warthin’s tumor specimens were detected by anti-TBET,Gata3,ROR-γt and Foxp3 antibodies,respectively.Antibody chip were applied to detect cytokine expression in 8 Warthin's tumors specimens.Results The percentage of positive cells of Th1,Th2,Th17 and Foxp^(3+)Treg were 2.48±2.129,0.70±1.101,0.97±1.002 and 6.26±3.567(±s,%).The number of Foxp^(3+)Treg cells was significantly higher than that of other lymphocyte subsets.Among the 40 cytokines,the cytokines related with chemokines,lymphatic homing,and lymphoid follicle formation expressed at a higher degree than the others,and the expression of the important senescence-associated secretory phenotype(SASP)factors such as TNF-α,IL-1βand IL-6 are also detected,and R language analysis showed that these SASP factors are strongly significantly correlated with the expression of inflammatory factors such as IL-8,IL-10,IL-12,GM-CSF,IL-11,IL-15 and IL-6sR.Conclusion Warthin’s tumor often occurs in elderly patients,and the distribution characteristics of lymphocyte subsets and cytokines may be related to the age-related changes of lymphointerstitial tissue.Therefore,it is speculated that age-related changes of lymphointerstitial tissue may play an important role in promoting the development of lymphointerstitial tissue.
