角茴香醇提颗粒对H1N1亚型流感病毒的体内抑制作用

摘要 为进一步开发针对流感病毒的治疗药物,本研究选用了H1N1亚型流感病毒感染小鼠模型,采用不同浓度的角茴香醇提颗粒给药治疗。通过评价小鼠的发病率、体重变化、存活率、肺指数、肺部病毒滴度、肺部病理变化、血细胞含量及细胞因子(TNF-α、IL-1β和IL-6)的表达水平,研究角茴香醇提颗粒对小鼠体内流感病毒的抑制作用和对流感病毒感染小鼠的治疗作用。结果发现,角茴香醇提颗粒能明显降低流感病毒感染小鼠的体重损失并提高小鼠的存活率。经过高浓度角茴香醇提颗粒治疗后,流感病毒感染小鼠的体重损失减少约10%,存活率提高60%。角茴香醇提颗粒能显著降低流感病毒感染小鼠的肺指数及肺部病毒滴度。经过高浓度角茴香醇提颗粒的治疗,在感染后第5天的小鼠肺指数从1.75%降低至1.21%,肺部病毒平均滴度从10^(4.65)TCID50/mL降低至10^(2.05)TCID50/mL。此外,角茴香醇提颗粒能明显缓解感染小鼠肺组织的病理损伤与炎症发展;能改善小鼠血液指标变化;能够对多种细胞因子的表达起到调节作用。由结果可知,角茴香醇提颗粒能明显抑制小鼠体内H1N1亚型流感病毒的活性,缓解肺部的急性肺损伤和炎症反应。所以说,中药角茴香是一种潜在的新型抗流感病毒药物。 In order to further develop therapeutic drugs against influenza virus,a mouse model of H1N1 subtype influenza virus infection was selected for this study,and different concentrations of alcoholic extracted particles of Hypecoum leptocarpum were used for treatment.The inhibitory effect of alcoholic extracted particles of H.leptocarpum on influenza virus in mice and its therapeutic effect on influenza virus-infected mice were investigated by evaluating the morbidity,body weight change,survival rate,lung index,lung virus titer,lung pathological changes,blood cell content and expression levels of cytokines(TNF-α,IL-1βand IL-6)in mice.The results showed that alcoholic extracted particles of H.leptocarpum significantly reduced the body weight loss and increased the survival rate of mice infected with influenza virus.After treatment with high concentrations of alcoholic extracted particles of H.leptocarpum,influenza virus-infected mice showed an approximately 10%reduction in body weight loss and a 60%increase in survival rate.Alcoholic extracted particles of H.leptocarpum significantly reduced the lung index and lung virus titer in influenza virus-infected mice.The lung index of mice at day 5 post-infection was reduced from 1.75%4.652.05 to 1.21%,and the lung mean virus titer was reduced from 10^(4.65) TCID_(50)/mL to 10^(2.05) TCID_(50)/mL after treatment with high concentrations of alcoholic extracted particles of H.leptocarpum.In addition,alcoholic extracted particles of H.leptocarpum could regulate the expression of various cytokines,resulting in significant alleviation of the pathological damage and inflammation development in the lung tissues of infected mice and improvement of the changes of blood parameters in mice.Therefore,it is clear that alcoholic extracted particles of H.leptocarpum can significantly inhibit the activity of H1N1 subtype of influenza virus in mice and alleviate the acute lung injury and inflammatory response in the lungs.The Chinese medicine H.leptocarpum is a potential novel anti-influenza virus drug.
