NaN_(3)诱变草莓突变体的表型筛选与SRAP分子鉴定

摘要 【目的】探索NaN_(3)对红花草莓的诱变效果,筛选出红花草莓突变体。【方法】在对NaN_(3)诱变红花草莓植株形态学初筛的基础上,进一步采用SRAP分子标记技术进行分子鉴定,通过聚类分析鉴定了红花草莓与其突变单株的遗传背景和亲缘关系。【结果】从M1代中筛选出23株具有不同表型变化的突变体,总突变率为4.6%,包括株型紧凑、株高变矮、茎纤细、小叶、叶片卷曲皱缩、花叶、白色花、三色花、花瓣短缩花、球型花、球形果、矩形果共12种突变性状;SRAP分子标记分析结果显示,8对SRAP引物扩增出的总条带数为72,多态性条带数为47,多态性比例为65%,23株表型突变株中有22个单株在DNA水平发生了基因突变;聚类分析表明,N-5、N-8和N-9与对照在遗传距离上相差最远,产生的变异最大,N-15与对照无差异。【结论】NaN_(3)诱变红花草莓育种是行之有效的,可产生草莓突变体,为红花草莓新品种选育以及相关研究提供参考依据。 【Objective】To study the mutagenic effect of NaN_(3)on safflower strawberry,to screen the mutants.【Method】Based on the morphological screening of NaN_(3)-induced safflower strawberry plants,SRAP molecular marker technology was further used for molecular identification.The genetic background and genetic relationship between safflower strawberry and its mutant plants were identified by cluster analysis.【Result】A total of 23 mutants with various phenotypic changes were screened from the M1 generation,and the total mutation rate was 4.6%,including 12 mutant traits such as compact plant type,short plant height,slender stem,small leaves,curled and wrinkled leaves,mosaic leaves,white flowers,tricolor flowers,petal shortening flowers,spherical flowers,spherical fruits and rectangular fruits.The results of SRAP molecular marker analysis showed that the total number of bands amplified by 8 pairs of SRAP primers was 72,the number of polymorphic bands was 47,and the polymorphic ratio was 65%.Cluster analysis showed that the genetic distance of N-5,N-8 and N-9 was the farthest from the control,and the variation was the greatest.【Conclusion】NaN_(3)mutagenesis of red-flowered strawberry breeding is effective and can produce strawberry mutants that provide a reference for the breeding of new red-flowered strawberry varieties and related research.
