摘要
骨关节炎(OA)引起的功能障碍是老年犬残疾的主要原因之一,旨在评估葛根素对前十字韧带切断(ACLT)诱导的犬OA模型中的抗炎和抗软骨降解作用。选取16只雄性德国牧羊犬,随机分为对照组、模型组、阳性对照组(200 mg/kg塞来昔布)、葛根素组(20 mg/kg),每组4只。对照组进行假手术,其他采用ACLT进行诱导OA模型,葛根素组和阳性对照组在造模2周后每天灌胃给药,干预至第8周。通过ELISA检测血清中IL-1β、IL-6、TNF-α、CTX-Ⅱ、COMP和CS 846,并在第8周时进行临床和影像学评估,综合评价葛根素对犬OA的保护作用。葛根素给药后能够显著抑制血清中IL-1β、IL-6和TNF-α的表达(P<0.05),其抗炎作用与口服塞来昔布相似,并且葛根素能够显著降低血清中骨代谢指标CTX-Ⅱ、COMP和CS 846的水平(P<0.05),在第8周时与模型组相比葛根素组临床评分和X线片评分均显著降低(P<0.05)。基于以上结果推测葛根素对犬OA模型的炎症反应有抑制作用,对软骨具有显著的保护作用。
Dysfunction caused by osteoarthritis(OA)is one of the main causes of disability in elderly dogs.This study aims to evaluate the anti-inflammatory and anti-cartilage degradation effects of puerarin on the canine OA model induced by anterior cruciate ligament transection(ACLT).Sixteen male German shepherd dogs were selected in the experiment and randomly divided into control group,model group,positive control group(200 mg/kg celecoxib),and puerarin-administered group(20 mg/kg),with four in each group.The ACLT method was used to make OA models,and the control group underwent sham operation,and the puerarin group and the positive control group were given daily intragastric administration for 2 weeks after modeling until the 8th week of the experiment.ELISA method was used to detect serum levels of interleukin-1β(IL-1β),interleukin 6(IL-6)and tumor necrosis factorα(TNF-α),and the C-terminal peptide of typeⅡcollagen(CTX-Ⅱ),cartilage oligomeric matrix protein(COMP)and chondroitin sulfate 846(CS846).Clinical and imaging evaluations were performed at the 8th week to comprehensively evaluate the protective effect of puerarin on canine OA.Puerarin significantly inhibited the expressions of IL-1β,IL-6 and TNF-αin serum within 8 weeks of administration(P<0.05),and its anti-inflammatory effect was similar to that of oral celecoxib.Puerarin reduced CTX in OA serum-levels ofⅡ,COMP and CS 846(P<0.05).In addition,after 8 weeks of puerarin treatment,clinical scores and X-ray scores were significantly reduced(P<0.05).Based on this,we speculate that puerarin has an inhibitory effect on the inflammatory response of the canine OA model and has a significant protective effect on cartilage.
