摘要
本研究采用不同方法从元宝枫籽油饼粕中提取蛋白并脱盐,以蛋白提取率、纯度和脱盐率为指标,选择最佳蛋白质提取工艺。不同的蛋白质提取方法中,采用Osborne法、盐析法、碱溶酸沉法、酸沉碱溶法的粕蛋白提取率和纯度分别为63.95%和73.12%、29.91%和87.74%、28.78%和84.52%、20.96%和54.59%,其中Osborne法的提取效果最好。提纯后,采用透析法对Osborne法提取的蛋白溶液进行脱盐研究,结果显示,粕蛋白溶液在4℃下透析3 h,脱盐率为38.8%,蛋白纯度为61.78%,脱盐效果和蛋白纯度综合最优。
To extract protein from Acer truncatum seed oil cake,the acid precipitation and alkali dissolution method,alkali dissolution and acid precipitation method,salting out method,and Osborne method were utilized in this work.With protein extraction rate,purity,and desalination rate as indexes,the best protein extraction method and desalination process were chosen.The extraction rate and purity of meal protein were 63.95%and 73.12%,29.91%and 87.74%,28.78%and 84.52%,20.96%and 54.59%,respectively,by the Osborne method,salting out method,alkali dissolution and acid precipitation method,acid precipitation and alkali disso-lution method.The Osborne method had the best extraction effect.Following purification,the protein solution extracted using the Osborne method was desalted using a dialysis bag.The protein solution was dialyzed at 4℃ for 3 h,the desalination rate was 38.8%,and the protein purity was 61.78%,which was determined to be the best desalination process.
