高糖高脂对人主动脉内皮细胞的影响及津力达联合通心络的干预作用

摘要 目的观察高糖高脂诱导人主动脉内皮细胞(HAECs)损伤的机制及津力达联合通心络的保护作用。方法采用高糖高脂诱导HAECs损伤,建立细胞损伤模型,将HAECs随机分为:空白(Normol)组、模型(Model)组、津力达(JLD)组、通心络(TXL)组、津力达联合通心络(JLD+TXL)组。检测各组细胞生存活性、细胞上清液内皮素(ET)-1、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性、细胞内活性氧簇(ROS)荧光强度、内皮型一氧化氮合酶(eNOS)蛋白表达。结果与Normal组相比,Model组细胞生存活性、细胞上清液中SOD活性、细胞eNOS蛋白表达显著下调,上清液ET-1、MDA含量明显上升,ROS荧光强度显著增强(P<0.01);与Model组相比,3个干预组细胞上清液ET-1含量显著下降,细胞eNOS蛋白表达明显上调(P<0.01),TXL和JLD+TXL组细胞生存活性、细胞上清液SOD活性显著上升,细胞ROS荧光强度和上清液MDA含量显著下降(P<0.05,P<0.01);与JLD组相比,TXL组和JLD+TXL组细胞上清液ET-1含量显著下降,细胞eNOS蛋白表达明显上调,细胞上清液SOD活性显著上升(P<0.01),JLD+TXL组细胞生存活性显著上升,细胞上清液MDA含量显著下降,细胞ROS荧光强度显著下降(P<0.01);与TXL组相比,JLD+TXL组细胞上清液ET-1含量显著下降,SOD活性显著上升,细胞eNOS蛋白表达明显上调(P<0.01)。结论高糖高脂可诱导HAECs功能损伤,津力达联合通心络能起到保护作用,其保护机制可能与减轻细胞氧化应激反应有关。 Objective To observe the mechanism of high glucose and high lipid induced human aortic endothelial cell(HAECs)injury and the protective effect of Jinlida combined with Tongxinluo.Methods HAECs damage was induced by high glucose and high lipid to establish cell damage model.HAECs were randomly divided into 5 groups:Normal group,Model group,Jinlida(JLD)group,Tongxinluo(TXL)group and Jinlida combined with Tongxinluo(JLD+TXL)group.The cell survival rate,the contents of endothelin(ET)-1 and malondialdehyde(MDA)in cell supernatant and the activity of superoxide dismutase(SOD),fluorescence intensity of intracellular reactive oxygen species(ROS)and protein expression of endothelial nitric oxide synthase(eNOS)were detected in each group.Results Compared with the Normal group,the cell survival activity,activity of SOD in cell supernatant and the protein expression of eNOS in cell were significantly decreased,the content of ET-1 and MDA in cell supernatant,and the fluorescence intensity of ROS were significantly increased in Model group(P<0.01).Compared with the Model group,the contents of ET-1 in cell supernatant of 3 intervention groups were significantly decreased and the protein expression of eNOS in cell was significantly increased(P<0.01),the cell survival activity and activity of SOD in cell supernatant were significantly increased,the cell fluorescence intensity of ROS and the content of MDA in cell supernatant were significantly decreased in TXL group and JLD+TXL group(P<0.05,P<0.01).Compared with the JLD group,the contents of ET-1 in cell supernatant were significantly decreased,the protein expression of eNOS in cell and the activity of SOD in cell supernatant were significantly increased in TXL group and JLD+TXL group(P<0.01);the cell survival activity was significantly increased,the content of MDA in cell supernatant and the fluorescence intensity of ROS in cell were significantly decreased in JLD+TXL group(P<0.01).Compared with the TXL group,the content of ET-1 in cell supernatant was significantly decreased,the activity of SOD and the protein expression of eNOS in cell were significantly increased in JLD+TXL group(P<0.01).Conclusions Jinlida combined with Tongxinluo could improve the functional damage of HAECs induced by high glucose and high lipid,and the protective mechanism might be related to reducing cellular oxidative stress response.
