酸性亮氨酸核磷酸蛋白32B对肺癌细胞增殖、迁移、凋亡的影响

Effect of ANP32B knockdown on proliferation,migration and apoptosis of lung cancer cells

目的探究酸性亮氨酸核磷酸蛋白32B(acidic leucine⁃rich nuclear phosphoprotein 32B,ANP32B)在肺癌(lung cancer,LC)中的作用机制。方法利用TCGA公共数据库,分析肺癌组织中ANP32B的表达情况。通过Kaplan⁃Meier Plotter生物信息数据库,检索分析ANP32B表达与患者预后的关系。通过体外细胞实验检测ANP32B在肺癌细胞及正常肺上皮细胞的表达差距。在肺癌细胞系中沉默ANP32B的表达水平,利用平板克隆实验计数细胞克隆团数目,利用划痕实验观察伤口愈合情况。Western blot检测ANP32B对通路蛋白的影响。结果通过TCGA数据库分析发现ANP32B在肺癌组织中表达水平高于癌旁组织,差异有统计学意义(P<0.001);通过Kaplan⁃Meier Plotter生物信息数据库发现ANP32B高表达的肺癌患者的总体生存期(overall survival,OS)较短(P<0.001)。通过体外细胞实验证明ANP32B在肺癌细胞的表达量高于正常肺上皮细胞。沉默ANP32B后,肺癌细胞增殖较对照组明显减慢(P<0.01),迁移能力明显下降(P<0.001)。ANP32B沉默后,凋亡相关蛋白Cleaved⁃Caspase3明显下调。结论ANP32B在肺癌组织及肺癌细胞系中高表达。沉默ANP32B表达水平后,可明显抑制A549细胞增殖、迁移。ANP32B可能抑制A549细胞的凋亡。

Objective To investigate the role and molecular mechanism of ANP32B in lung cancer(LC).Methods The mRNA expression of ANP32B in lung cancer was analyzed using theTCGA public database.Through Kaplan⁃Meier Plotter bioinformatics database,the relationship between ANP32B expression and patient prognosis was retrieved and analyzed.The expression gap of ANP32B in lung cancer cells and normal lung epithelial cells was determined by in vitro cell assay.The expression level of ANP32B was silenced in lung cancer cell lines,the number of cell clusters was counted using the plate cloning assay,and wound healing was observed using the scratch assay.Results The expression level of ANP32B in lung cancer tissue was higher than that in paracancerous tissue according to TCGA database analysis,with statistical significance(P<0.001);The Kaplan⁃Meier Plotter⁃bioinformatics database found that the overall survival(OS)of lung cancer patients with high ANP32B expression was shorter.The expression level of ANP32B was silenced in lung cancer cell lines,the number of cell clusters was counted using the plate cloning assay,and wound healing was observed using the scratch assay.Conclusions ANP32B is highly expressed in lung cancer tissues and lung cancer cell lines.Silencing ANP32B expression levels significantly inhibited the proliferation and migration of A549 cells.ANP32B may inhibit the apoptosis of A549 cells.
