摘要
丹参(Salvia miltiorrhiza Bge.)是我国传统大宗中药材,探究丹参有效成分含量的杂种优势,可为提高丹参的产量、品质和促进丹参新品种选育提供基础。将川丹参(BH)和山东丹参(SD)进行杂交,得到正反交杂交F_(1)代,采用高效液相色谱(HPLC)法测定其有效成分含量,并运用SPSS软件进行主成分分析和单因素方差分析。结果表明:正交F_(1)代(BH×SD)的迷迭香酸和丹酚酸B的含量分别为3.40 mg/g和75.35 mg/g,隐丹参酮、丹参酮I和丹参酮ⅡA的含量分别为5.41 mg/g、1.76 mg/g和5.48 mg/g。反交F_(1)代(SD×BH)的迷迭香酸和丹酚酸B的含量分别为3.28 mg/g、63.38 mg/g,隐丹参酮、丹参酮I和丹参酮ⅡA的含量分别为4.6 mg/g、1.74 mg/g和5.00 mg/g。BH×SD和SD×BH的丹酚酸B含量显著高于两亲本,是中国药典规定含量的2倍以上,总丹参酮含量分别为12.65 mg/g和11.34 mg/g,高出中国药典规定含量的406%和354%。BH与SD杂交后所得到的F_(1)代在有效成分上表现出不同程度的杂种优势,可以作为新品种选育的种质资源。
Salvia miltiorrhiza Bge.is a traditional Chinese herbal medicine.Exploring the heterosis of effective components content of Salvia miltiorrhiza Bge.can provide a basis for improving the yield,quality and promoting the breeding of new Salvia miltiorrhiza Bge.varieties.In this study,Salvia miltiorrhiza Bge.from Sichuan(BH)and from Shandong(SD)were crossed to obtain the F_(1) generation,and their contents of active ingredients were compared by high performance liquid chromatography(HPLC),followed by principal component analysis and one-way ANOVA using SPSS software.In the orthogonal F_(1) generation(BH×SD),the contents of rosmarinic acid and salvianolic acid B were 3.40 mg/g and 75.35 mg/g,and the contents of cryptotanshinone,tanshinone I and tanshinone IIA were 5.41 mg/g,1.76 mg/g and 5.48 mg/g.In the reverse crossed F_(1) generation(SD×BH),the contents of rosmarinic acid and salvianolic acid B were 3.28 mg/g and 63.38 mg/g,and the contents of cryptotanshinone,tanshinone I and tanshinone IIA were 4.6 mg/g,1.74 mg/g and 5.00 mg/g.The content of salvianolic acid B in BH×SD and SD×BH was significantly higher than that of the two parents,which was more than twice the content of stipulated in the Chinese Pharmacopoeia.And the total tanshinone contents were 12.65 mg/g and 11.34 mg/g,which were 406%and 354%higher than those specified in the Chinese Pharmacopoeia.The F_(1) generation obtained by crossing BH and SD showed different degrees of heterosis in effective components,which could be used as germplasm resources for new variety breeding.
