糖肾清颗粒对糖尿病肾病大鼠足细胞Synaptopodin表达的影响研究

摘要 目的:研究糖肾清颗粒对糖尿病肾病模型大鼠足细胞Synaptopodin表达的影响,探讨其对糖尿病肾病模型大鼠肾脏的保护作用。方法:选取60只雄性Wistar大鼠,随机分为模型组(给予生理盐水)、空白组(给予生理盐水)、贝那普利组[给予盐酸贝那普利0.9 mg/(kg·d)]、糖肾清高剂量组[给予糖肾清颗粒24.3 mg/(kg·d)]、糖肾清低剂量组[给予糖肾清颗粒12.15 mg/(kg·d)],采用右肾摘除法加腹腔注射链脲佐菌素法建立糖尿病肾病大鼠模型。给药12周后,检测各组24 h尿蛋白定量、尿N-乙酰-β-D-葡萄糖苷酶(NAG)、总胆固醇(TC)、甘油三酯(TG)、空腹血糖(FBG)、Synaptopodin水平。结果:模型组、贝那普利组、糖肾清低剂量组、糖肾清高剂量组尿NAG、24 h尿蛋白定量、TC、TG、FBG水平均高于空白组,Synaptopodin水平低于空白组,差异有统计学意义(P<0.05);尿NAG、24 h尿蛋白定量、TC、TG、FBG在其他四组中的水平为模型组>贝那普利组>糖肾清低剂量组>糖肾清高剂量组,Synaptopodin水平为模型组<贝那普利组<糖肾清低剂量组<糖肾清高剂量组,差异均有统计学意义(P<0.05)。结论:糖肾清颗粒能显著降低糖尿病肾病模型大鼠血糖、血脂、24 h尿蛋白定量、尿NAG水平,上调Synaptopodin在肾组织中的表达,保护肾功能。 Objective:To study the effect of Tangshenqing granules on Synaptopodin in podocytes of diabetic nephropathy model rats,and to explore the protective effect on the kidney of diabetic nephropathy model rats.Methods:Sixty male Wistar rats were selected and randomly divided into model group(giving physiological saline),blank group(giving physiological saline),benazepril group[giving benazepril hydrochloride 0.9 mg/(kg·d)],Tangshenqing high-dose group[giving Tangshenqing granules 24.3 mg/(kg·d)],and Tangshenqing low-dose group[giving Tangshenqing granules 12.15 mg/(kg·d)].The rat model of diabetic nephropathy was established by right kidney excision and intraperitoneal injection of streptozotocin.After 12 weeks of administration,24 h urinary protein quantification,urinary N-Acetyl-β-D-glucosidase(NAG),total cholesterol(TC),triglyceride(TG),fasting blood glucose(FBG)and Synaptopodin level in each group were detected.Results:The urinary NAG,24 h urinary protein quantification,TC,TG and FBG levels in the model group,benazepril group,Tangshenqing low-dose group and Tangshenqing high-dose group were higher than those in the blank group,and the Synaptopodin levels were lower than those in the blank group,and the differences were statistically significant(P<0.05).The levels of urinary NAG,24 h urinary protein quantification,TC,TG and FBG in the other four groups were as follows:model group>benazepril group>Tangshenqing low-dose group>Tangshenqing high-dose group,and the levels of Synaptopodin were as follows:model group<benazepril group<Tangshenqing low-dose group<Tangshenqing high-dose group,and the differences were statistically significant(P<0.05).Conclusion:Tangshenqing granules can significantly reduce blood glucose,blood lipids,24 h urinary protein quantification,urinary NAG,up-regulate the expression of Synaptopodin in renal tissue,and protect renal function in diabetic nephropathy model rats.
