摘要
目的研究补肾活血方对免疫性血小板减少症(ITP)模型小鼠白细胞介素(IL)-6、IL-17、IL-21、IL-22、肿瘤坏死因子α(TNF-α)等免疫因子及骨髓巨核细胞的影响。方法建立小鼠ITP模型,随机分为正常组、模型组、泼尼松组、补肾活血高、中、低剂量组。相应干预措施持续14 d。采用ELISA法测定检测各组小鼠IL-6、IL-17、IL-21、IL-22、TNF-α含量,并取股骨骨髓测定骨髓巨核细胞数量。结果与正常组相比,ITP模型组小鼠骨髓巨核细胞数量明显增多,产板巨核细胞数量显著下降,IL-6、IL-17A、IL-21、IL-22、TNF-α水平明显上调,差异有统计学意义(P<0.01)。补肾活血方高剂量组小鼠巨核细胞数量显著低于模型组(P<0.05),产板巨核细胞数差异无统计学意义,但有一定回升。IL-6、IL-17A、IL-21、IL-22、TNF-α水平补肾活血方高剂量组与模型组相比显著下调(P<0.01),补肾活血方中剂量组与模型组相比差异有统计学意义(P<0.05)。结论补肾活血方能通过下调ITP小鼠模型体内IL-6、IL-17A、IL-21、IL-22、TNF-α水平,一定程度改善巨核细胞成熟障碍,提高血小板数量。
Objective To study the effects of Bushen Huoxue Prescription(补肾活血方)on immune factors such as interleukin(IL)-6,IL-17,IL-21,IL-22,tumor necrosis factor-α(TNF-α)and bone marrow megakaryocytes in immune thrombocytopenia(ITP)model mice.Methods Mouse ITP models were established and randomly divided into normal group,model group,prednisone group,Bushen Huoxue Prescription high,medium and low dose groups.The corresponding intervention lasted for 14 days.The contents of IL-6,IL-17,IL-21,IL-22 and TNF-αwere detected by ELISA,and the number of bone marrow megakaryocytes was determined by femoral bone marrow.Results Compared with those of the normal group,the number of bone marrow megakaryocytes in the ITP model group was significantly increased,the number of production plate megakaryocytes was significantly decreased,and the contents of IL-6,IL-17A,IL-21,IL-22 and TNF-αwere significantly up-regulated(P<0.01).The number of megakaryocytes in Bushen Huoxue Prescription high dose group was significantly lower than that in the model group(P<0.05),and there was no significant difference in the number of megakaryocytes in production plate,but there was a certain increase.The contents of IL-6,IL-17A,IL-21,IL-22 and TNF-αin the Bushen Huoxue Prescription high dose group were significantly down-regulated compared with those in the model group(P<0.01),and the Bushen Huoxue Prescription high dose group was also significantly different from those in the model group(P<0.05).Conclusion Bushen Huoxue Prescription can promote the maturation disorder of bone marrow megakarocytes and increase the number of platelets by down-regulating the levels of IL-6,IL-17A,IL-21,IL-22 and TNF-αin ITP mouse model.
