低共熔溶剂对虎杖中虎杖苷的高效绿色提取工艺优化研究 被引量:2

Optimization of Green Extraction Process of Polydatin from Huzhang(Polygoni Cuspidati Rhizoma Et Radix)by Deep Eutectic Solvent

摘要 目的建立虎杖中虎杖苷绿色高效提取方法。方法以中药虎杖粉末为原料,虎杖苷提取率为指标,优选的低共熔溶剂(deep eutectic solvent,DES)为提取溶液,对DES的摩尔比和含水量,以及提取工艺(温度、时间、固液比)进行参数优化。结果氯化胆碱-乙二醇(choline chloride-ethylene glycol,ChCl-Gly),氯化胆碱-乳酸(choline chloride-lactic acid,ChCl-Lac),甜菜碱-乳酸(betaine-lactic acid,Bet-Lac)DES是适宜的绿色提取溶剂,其最佳提取参数分别为:氯化胆碱-乙二醇溶液摩尔比为1∶4,含水量为40%(v/v,下同),固液比为1∶10(g/mL,下同),搅拌温度为80℃,搅拌时间为45 min;氯化胆碱-乳酸溶液,摩尔比为1∶3,含水量为40%,固液比为1∶20,搅拌温度为40℃,搅拌时间为30 min;甜菜碱-乳酸溶液,摩尔比为1∶4,含水量为60%,固液比为1∶20,搅拌温度为40℃,搅拌时间为30 min,虎杖苷的提取含量相比于《中华人民共和国药典》2020版提取方法有极大地提升。结论氯化胆碱-乙二醇、氯化胆碱-乳酸、甜菜碱-乳酸等低共熔溶剂可以替代传统的稀乙醇作为虎杖中虎杖苷的高效绿色提取溶剂,且该方法操作简便,用时短,效率高。 Objective To establish a efficient and green method for extraction of polydatin from Huzhang(Polygoni Cuspidati Rhizoma Et Radix).Methods Using Huzhang(Polygoni Cuspidati Rhizoma Et Radix)powder as extraction material,the yield of polydatin was selected as the index,and the optimal deep eutectic solvent(DES)was selected as the extraction solution.The molar ratio,water content and extraction process parameters(temperature,time,solid-liquid ratio)of DES were optimized.Results The experiment results showed that choline chloride-ethylene glycol(ChCl-Gly),choline chloride-lactic acid(ChCl-Lac)and betaine-lactic acid(Bet-Lac)were a suitable green extraction solvent for extraction polydatin.The optimized extraction parameter was followed:the molar ratio of choline chloride-ethylene glycol(ChCl-Gly)solution was 1∶4,the water content was 40%(v/v,same blow),the solid-liquid ratio was 1∶10(g/mL,same blow),the stirring temperature was 80℃,and the stirring time was 45 min.The molar ratio of choline chloride-lactic acid was 1∶3,the water content was 40%,the solid-liquid ratio was 1∶20,the stirring temperature was 40℃and the stirring time was 30 min.Betaine-lactic acid solution,the molar ratio of 1∶4,the water content of 60%,the solid-liquid ratio of 1∶20,the stirring temperature of 40℃,the stirring time of 30 min,compared with the extraction method of Pharmacopoeia of the People's Republic of China 2020 edition,the extraction content of polydatin was greatly improved.Conclusion DES such as choline chlorideethylene glycol,choline chloride-lactic acid and betaine-lactic acid may replace the traditional ethanol as the efficient green extraction solvent of polydatin from Huzhang(Polygoni Cuspidati Rhizoma Et Radix),and this method is simple,timeless and high efficiency.
