环状RNA UBAP2和环状RNA VRK1在子痫前期患者血清中的表达及其与妊娠结局的关系

Expression of circular RNA UBAP2 and circular RNA VRK1 in serum of patients with preeclampsiaand their relationship with pregnancy outcome

目的探讨环状RNA UBAP2(circUBAP2)、环状RNA VRK1(circVRK1)在子痫前期(PE)患者血清中的表达水平及其与妊娠结局的关系。方法选取2019年10月至2022年8月于石家庄市第四医院分娩的134例PE患者作为PE组,另选取同期分娩的134例健康孕妇作为健康组。比较PE组、健康组血清circUBAP2、circVRK1表达水平及不良妊娠结局发生率。依据PE患者妊娠结局,将其分为妊娠良好组(n=94)和妊娠不良组(n=40);比较血清circUBAP2、circVRK1表达水平;采用受试者工作特征(ROC)曲线分析血清circUBAP2、circVRK1预测PE患者妊娠结局的价值;采用Logistic回归分析PE患者妊娠结局的影响因素。结果PE组患者血清circUBAP2表达水平低于健康组(P<0.05),circVRK1表达水平及总不良妊娠结局发生率高于健康组(P<0.05)。妊娠不良组PE患者收缩压、重度PE占比、血清circVRK1表达水平高于妊娠良好组(P<0.05)。血清circUBAP2、circVRK1预测PE患者妊娠结局的曲线下面积(AUC)分别为0.869、0.686,截断值分别为0.39、2.50,灵敏度分别为77.5%、70.0%,特异度分别为84.0%、63.8%。重度PE、低表达circUBAP2、高表达circVRK1与PE患者妊娠不良结局密切相关(P<0.05)。结论PE患者血清circUBAP2表达水平较低,circVRK1表达水平较高,与PE患者妊娠结局密切相关,且circUBAP2或可成为临床预测PE患者妊娠结局的辅助指标。

Objective To investigate the expression levels of circular RNA UBAP2(eireUBAP2)and circular RNA VRK1(cireVRK1)in the serum of patients with preeclampsia(PE)and their relationship with pregnancy outcome.Methods A total of 134 PE patients delivered in Shjiazbuang Fourth Hospital from October 2019 to August 2022 were regarded as PE group,another 134 healthy pregnant women who delivered at the same time were included as the healthy group.The expression levels of circUBAP2 and circVRK1 in serum and the incidence of adverse pregnancy outcome were compared between PE group and healthy group.According to the pregnancy outcome of PE patients,they were divided into good pregnancy group(n=94)and poor pregnancy group(n=40),the expression levels of circUBAP2 and cireVRK1 in serum were compared;receiver operator characteristic(ROC)curve was performed to analyze the value of serum circUBAP2 and cireVRK1 in predic ting pregnancy outcome of PE patients;Logistic regression analysis was used to analyze the influencing factors of pregnancy outcome in PE patients.Results The expression level of cirpUBAP2 in PE group was lower than that in healthy group(P<0.05),the expression level of circVRK1 and the incidence of adverse pregnancy outcome were higher than those in the healthy group(P<0.05).The systolic blood pressure,the proportion of severe PE,and the expression level of cireVRK1 in serum of PE patients in poor pregnancy group were higher than those in good pregnancy group(P<0.05).The area under the curve(AUC)of cireUBAP2 and cireVRK1 in serum to predict pregnancy outcome of PE patients was 0.869 and 0.686,respectively,with cut-off value of 0.39 and 2.50,respectively,with sensitivity of 77.5%and 70.0%,respectively,and specificity of 84.0%and 63.8%,respectively.Severe PE,low expres sion of circUBAP2 and high expression of circVRK1 were closely related to adverse pregnancy outcomes in PE patients(P<0.05).Conclusions The expression level of circUBAP2 in serum of PE patients is low,and the expression level of cireVRK1 is high,which is closely related to the pregnancy outcome of PE patients,and circUBAP2 may be an auxiliary index for clinical prediction of pregnancy outcome of PE patients.
