摘要 骨代谢和稳态以骨重塑能力维持平衡,老年人骨质疏松常伴有骨重塑异常,即骨吸收大于骨形成。骨质疏松是常见的代谢性骨疾病之一,随着人口老龄化,其发病率急剧增加。哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)是一种进化保守的丝氨酸/苏氨酸激酶,它主要是通过控制细胞过程(如蛋白质合成、代谢和自噬等)调节机体生长和平衡。近年来,围绕“mTOR-骨代谢”的研究较为火热,因mTOR对骨形成或骨吸收的作用报道不一,本文着重对这一领域的研究结果作一综述,旨在为骨质疏松症的治疗寻找新的药物靶点和治疗方向。 Bone metabolism and homeostasis are balanced by bone remodeling.Osteoporosis in the elderly is often accompanied by abnormal bone remodeling,that is,bone resorption is greater than bone formation.Osteoporosis is one of the most common metabolic bone diseases,of which the incidence is increasing rapidly,with the aging of the population.Mammalian target of rapamyci(mTOR)is an evolutionarily conserved serine/threonine kinase.It mainly regulates the growth and balance of the body by controlling cellular processes(such as protein synthesis,metabolism and autophagy).In recent years,the research about the relationship between mTOR and bone metabolism is hot.Due to different reports on the effects of mTOR on bone formation or bone resorption,this paper focuses on this field,in order to find new drug targets and treatment directions for osteoporosis.
