高效液相色谱法同时测定干姜配方颗粒中5种成分含量 被引量:3

摘要 目的建立同时测定干姜配方颗粒中5种成分含量的高效液相色谱法。方法色谱柱为Thermo Syncronis C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(梯度洗脱),流速为1.0 mL/min,检测波长为280 nm,柱温为30℃,进样量为20μL。结果姜酮、6-姜酚、8-姜酚、6-姜烯酚、10-姜酚的质量浓度分别在0.5080~50.80μg/mL(r=0.9991)、2.0150~201.50μg/mL(r=0.9999)、0.5160~51.60μg/mL(r=0.9998)、0.6030~60.30μg/mL(r=0.9997)、0.5210~52.10μg/mL(r=0.9993)范围内与峰面积线性关系良好;精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均加样回收率分别为96.21%,99.64%,98.85%,98.15%,98.08%,RSD分别为1.61%,1.15%,1.34%,1.18%,1.28%(n=6)。结论所建立的方法操作简便,结果准确,重复性好,可用于同时测定干姜配方颗粒中5种成分的含量。 Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of five components in Ganjiang Formula Granules.Methods The chromatographic column was Thermo Synronis C_(18)column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-water(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 280 nm,the column temperature was 30℃,and the injection volume was 20μL.Results The linear ranges of zingerone,6-gingerol,8-gingerol,6-shogaol,10-gingerol were 0.5080-50.80μg/mL(r=0.9991),2.0150-201.50μg/mL(r=0.9999),0.5160-51.60μg/mL(r=0.9998),0.6030-60.30μg/mL(r=0.9997)and 0.5210-52.10μg/mL(r=0.9993)respectively.The RSDs of precision,stability and repeatability tests were all lower than 2.0%.The average recovery rates of the above five components were 96.21%,99.64%,98.85%,98.15%and 98.08%respectively,with RSDs of 1.61%,1.15%,1.34%,1.18%and 1.28%respectively(n=6).Conclusion The established method is sample,accurate and repeatable,which can be used for the simultaneous determination of five components in Ganjiang Formula Granules.
