摘要
以杂交墨兰无菌播种形成的根状茎为试验材料,比较不同基本培养基、植物生长调节剂和外界环境等要素对墨兰增殖、叶芽分化和花芽分化的影响。结果表明:花宝1号+花宝2号有利于根状茎的生长与增殖,生长速度达4.43;高浓度6-BA有利于根状茎的生长与增殖,增殖系数达5.90。在光照强度1400~1500 lx下,适宜根状茎叶芽分化的培养基为MS+5 mg/L NAA+1.0 mg/L 6-BA+0.5 mg/L TDZ+40 g/L Su+7.0 g/L Ag,叶芽分化率达86.67%。通过调节温差,进行低温处理,有利于墨兰根状茎花芽分化。
The rhizome formed by aseptic seeding of hybrid Cymbidium sinense was used as the test material to compare the effects of different basic media,plant growth regulators,and external environment on Cymbidium sinense proliferation,leaf bud differentiation,and flower bud differentiation.The results showed that:HyponexⅠ+HyponexⅡwas beneficial to the growth and proliferation of rhizomes,with a growth rate of 4.43;A high concentration of 6-BA was beneficial to the growth and proliferation of rhizomes,with a proliferation coefficient of 5.90.Under the light intensity of 1400~15001x,the suitable medium for rhizome and leaf bud differentiation is MS+5 mg/L NAA+1.0 mg/L 6-BA+0.5 mg/L TDZ+40 g/L Su+7.0 g/L Ag,the leaf bud differentiation rate reached 86.67%.By adjusting the temperature difference and performing low-temperature treatment,there is conducive to the differentiation of flower buds of Cymbidium sinense rhizome.
