大豆皂苷Aa联合有氧运动对肥胖大鼠的减脂效果

Lipid-reducing Effect of Soyasaponin Aa Combined with Aerobic Exercise on Obese Rats

摘要 本研究探讨大豆皂苷Aa(Soyasaponin Aa,SS Aa)联合有氧运动对肥胖大鼠的减脂效果及机制。将大鼠分为CON组、HFD组、HFD+AE组、HFD+SS Aa组、HFD+AE+SS Aa组,每组12只大鼠。CON组大鼠使用正常饲料喂养,其他组大鼠使用高脂饲料喂养。HFD+AE组和HFD+AE+SS Aa组大鼠进行6周有氧运动。HFD+SS Aa组和HFD+AE+SS Aa组大鼠在有氧运动前1 h灌胃2 mL 20 mg/kg的大豆皂苷Aa溶液,共灌胃6周。检测了各组大鼠的体重、血清脂质代谢指标(TC,TG,LDL-c,HDL-c,FFA)、肝脏抗氧化指标(SOD,CAT和MDA)和炎症指标(TNF-α,IL-1β和IL-6)水平。通过RT-qPCR检测肝脏C/EBPα、Adiponectin、SREBP1c、a P2、FAS和Resistin m RNA水平。通过Western blot检测肝脏中细胞核Nrf2蛋白表达和NF-κB p65的磷酸化水平。与HFD+AE组和HFD+SS Aa组比较,HFD+AE+SS Aa组大鼠的体重降低(P<0.05);血清TC、TG、LDL-c和FFA水平降低,HDL-c水平升高(P<0.05);肝脏SOD和CAT水平升高,MDA水平降低(P<0.05);肝脏TNF-α、IL-1β和IL-6水平降低(P<0.05);肝脏C/EBPα、Adiponectin、SREBP1c、aP2、FAS和Resistin mRNA水平降低(P<0.05);肝脏中细胞核Nrf2蛋白表达水平升高,NF-κB p65的磷酸化水平降低(P<0.05)。大豆皂苷Aa联合有氧运动有效降低了肥胖大鼠的体重,纠正了脂质代谢紊乱和抗氧化系统失衡,抑制了慢性炎症,其机制与调节脂质代谢基因转录、Nrf2和NF-κB的活化有关。 To explore the fat-reducing effect and mechanism of Soyasaponin Aa(SS Aa)combined with aerobic exercise on obese rats.Rats were divided into CON group,HFD group,HFD+AE group,HFD+SS Aa group and HFD+AE+SS Aa group,with 12 rats in each group.Rats in CON group were fed with normal diet,while rats in other groups were fed with high-fat diet.The rats in HFD+AE group and HFD+AE+SS Aa group were given aerobic exercise for 6 weeks.The rats in HFD+SS Aa group and HFD+AE+SS Aa group were intragastrically perfused with 2 mL 20 mg/kg soyasaponin Aa solution 1 hour before aerobic exercise for 6 weeks.The body weight,serum lipid metabolism indexes(TC,TG,LDL-c,HDL-c,FFA),liver antioxidant indexes(SOD,CAT and MDA)and inflammatory indexes(TNF-α,IL-1βand IL-6)were measured.The levels of C/EBPα,adiponectin,SREBP1c,aP2,FAS and resistin mRNA in liver were detected by RT-qPCR.The expression of nuclear Nrf2 protein and the phos phorylation level of NF-kb p65 in liver were detected by Western blot.Compared with HFD+AE group and HFD+SS Aa group,the body weight of rats in HFD+AE+SS Aa group decreased(P<0.05),the levels of serum TC,TG,LDL-c and FFA decreased and the level of HDL-c increased(P<0.05),the levels of SOD and CAT in liver increased and the level of MDA decreased(P<0.05),the levels of TNF-α,IL-1βand IL-6 in liver decreased(P<0.05),the levels ofC/EBPα,adiponectin,SREBP1c,aP2,FAS and sesistin mRNA in liver decreased(P<0.05),the expres-sion of nuclear Nrf2 protein in liver increased and the phosphorylation level of NF-kB p65 decreased(P<0.05).Soyasaponin Aa combined with aerobic exercise can effectively reduce the body weight of obese rats,correct the disorder of lipid metabolism and the imbalance of antioxidant system,and inhibit chronic inflammation.The mechanism is related to the regulation of lipid metabolism gene transcription and the activation of Nrf2 and NF-kB.
