海藻活性成分岩藻甾醇联合运动改善肥胖大鼠脂质代谢

Algae Active Ingredient Fucosterol Combined with Exercise Improves Lipid Metabolism in Obese Rats

摘要 甾醇是功能性食品的重要组成部分。岩藻甾醇(FUC)是海藻中的主要甾醇,具有多种食用和药用价值。为了揭示海藻活性成分岩藻甾醇联合运动对肥胖大鼠的脂质代谢、抗氧化系统和慢性炎症的影响。本实验将大鼠组,对照组使用正常饲料喂养,并进行运动处理,跑台运动和给药时间均为6周。实验后,检测了各组大鼠的体重、血清脂质代谢指标(TC,TG,LDL-c,HDL-c,FFA)、肝脏氧化应激指标(SOD,CAT和MDA)和炎症指标(TNF-α,IL-1β和IL-6)水平。通过RT-qPCR检测肝脏C/EBPα、Adiponectin、SREBP1c、aP2、FAS和Resistin mRNA水平。通过Western blot检测肝脏中细胞核Nrf2蛋白表达和NF-κB p65的磷酸化水平。结果显示,与HFD+E组和HFD+FUC组比较,HFD+E+FUC组大鼠的体重降低(P<0.05);血清TC、TG、LDL-c和FFA水平降低,HDL-c水平升高(P<0.05);肝脏SOD和CAT水平升高,MDA水平降低(P<0.05);肝脏TNF-α、IL-1β和IL-6水平降低(P<0.05);肝脏C/EBPα、Adiponectin、SREBP1c、aP2、FAS和Resistin mRNA水平降低(P<0.05);肝脏中细胞核Nrf2蛋白表达水平升高,NF-κB p65的磷酸化水平降低(P<0.05)。本实验表明,岩藻甾醇联合运动可最大程度的纠正肥胖大鼠脂质代谢紊乱和抗氧化系统失衡,并抑制慢性炎症,其机制与调节脂质代谢基因转录、Nrf2和NF-κB的活化有关。本实验认为岩藻甾醇可能是来源于海藻的一种优良的运动补充剂。 Sterol is an important part of functional food.Fucosterol(FUC)is the main sterol in seaweed,which has a variety of edible and medicinal values.The purpose of this study was to reveal the effects of fucosterol on lipid metabolism,antioxidant system and chronic inflammation in obese rats.In this experiment,rats were grouped,the control group was fed with normal diet,and the treating groups were fed with high fat diet.The time of treadmill exercise and administration was 6 weeks.After the experiment,the body weight,serum lipid metabolism indexes(TC,TG,LDL-c,HDL-c,FFA),liver oxidative stress indexes(SOD,CAT and MDA)and inflammatory indexes(TNF-α,IL-1βand IL-6)were measured.The levels ofC/EBPα,Adiponectin,SREBP1c,aP2,FAS and Resistin mRNA in liver were detected by RT-qPCR.The expression of nuclear Nrf2 protein and the phosphorylation level of NF-kB p65 in liver were detected by Western blot.The results showed that compared with HFD+E group and HFD+FUC group,the body weight ofrats in HFD+E+FUC group decreased,the levels of serum TC,TG,LDL-c and FFA decreased and the level of HDL-c increased,the levels ofSOD and CAT in liver increased and the level of MDA decreased,the levels of TNF-α,IL-1βand IL-6 in liver decreased,and the levels ofC/EBPα,Adiponectin,SREBPlc,aP2,FAS and Resistin mRNA in liver decreased,the expression of nuclear Nrf2 protein in liver increased and the phosphorylation level ofNF-kB p65 decreased(P<0.05).This experiment shows that fucosterol com-bined exercise can correct the disorder of lipid metabolism and the imbalance of antioxidant system in obese rats to the greatest extent,and inhibit chronic inflammation,and its mechanism is related to the regulation of lipid metabolism gene transcription and the activation of Nrf2 and NF-kB.It is suggested that fucosterol may be an excellent exercise supplement from seaweed.
