摘要
试验旨在研究提取条件对黑果腺肋花楸果原花青素、花色苷提取量的影响及其与牛磺胆酸钠的结合能力。试验以乙醇水溶液为溶剂同时提取黑果腺肋花楸果中原花青素和花色苷,以原花青素、花色苷提取量为考察指标,通过单因素试验和正交试验优化提取工艺。结果显示,在提取溶剂为50%乙醇水溶液、温度为80℃、时间为1.5 h、液料比为65 mL/g、提取2次的优化条件下,原花青素提取量为10.37 mg/g,花色苷提取量为0.88 mg/g,总提取量为11.25 mg/g,出膏率29.90%。黑果腺肋花楸果提取物与牛磺胆酸钠结合率IC50值为0.83 g/L。研究表明,采用50%乙醇水溶液同时提取黑果腺肋花楸果原花青素、花色苷,优化后的提取效果良好,工艺稳定,其提取物具有与牛磺胆酸钠结合的能力,结合能力随提取物浓度的增加逐渐增强。
The study was to study the effect of anthocyanins and proanthocyanidins on the amount of extract and its binding ability to natrium taurocholicum.In the experiment,the proanthocyanidins and anthocyanins of Aronia melanocarpa were simultaneously extracted by ethanol aqueous solution,with the extraction amount of proanthocyanidins and anthocyanins as testing factor,the extraction process was optimized by univariate test and orthogonal test.The results showed that under the optimized conditions of extraction solvent of 50%ethanol solution,temperature of 80℃,time of 1.5 h,liquid to solid ratio of 65 mL/g and extraction twice,the yield of proanthocyanidins and anthocyanins were 10.37 mg/g and 0.88 mg/g,the total yield and the cream yield were 11.25 mg/g and 29.90%.The ICso value of the binding rate of the extract of Aronia melanocarpa with taurocholate was 0.83 g/L.The study indicates with 50%ethanol water solution,the anthocyanins and proanthocyanidins are extracted simultaneously,which is good after orthogonal optimization,and the process was stable.The extracts have the ability to combine with natrium taurocholicum,and the binding is gradually enhanced with the increase of extract concentration.
