摘要
为提高脱毒马铃薯试管薯生产效率,本文开展了马铃薯试管薯诱导培养基及诱导技术的优化试验。结果表明,改良壮苗培养基配方(MS+活性炭10 g/L+蔗糖50 g/L+椰子汁100 m L/L)较常规培养基配方(MS+蔗糖30 g/L+活性炭5 g/L)培养的马铃薯组培苗平均茎粗增加了42%,平均叶面积增加了60%;将壮苗培养后的组培苗暗培养时间改为10 d,然后散射光培养10 d,随后继续暗培养,改良条件下诱导试管薯平均单株数量为4.2个,较对照增加35.8%;平均单薯质量为319 mg,较对照增加10.9%。本文为脱毒马铃薯试管薯的生产提供了新的方法。
In order to improve the production efficiency of virus-free potato in vitro tubers,this article conducted optimization experiments on potato in vitro tuber induction medium and induction technology.The results showed that the improved strong seedling culture medium formula(MS+activated carbon 10 g/L+sucrose 50 g/L+coconut juice 100 mL/L)increased the average stem diameter of potato tissue culture seedlings by 42%and the average leaf area by 60%compared to the conventional medium formula(MS+sucrose 30 g/L+activated carbon 5 g/L).Change the dark culture time of the tissue cultured seedlings after strong seedling cultivation to 10 days,then scatter ligh culture for 10 days,and continue dark culture.Under the improved conditions,the average number of induced test tube potatoes per plant was 4.2,an increase of 35.8%compared to the control.The average weight of a single potato was 319 mg,an increase of 10.9%compared to the control.This article provided a new method for the production of virus-free potato test tube potatoes.
