摘要
以夏橙为原料,采用纤维素酶和木聚糖酶共同酶解,利用水蒸气蒸馏法从橙皮中提取精油。通过单因素实验得出混合酶较适宜的酶解温度、酶解pH及酶解时间;在单因素实验的基础上,采用响应面法探究两种酶混合辅助水蒸气蒸馏法提取橙皮精油的最佳工艺条件。结果表明:纤维素酶的适宜添加量为橙皮质量的1.2%,木聚糖酶的适宜添加量为橙皮质量的2.2%;混合酶辅助水蒸气蒸馏法提取橙皮精油的最佳工艺条件为酶解温度为41℃、酶解pH=6.337、酶解时间为1.4 h,所得橙皮精油的提取率为1.54%,比未添加酶时的精油提取率高出1.02个百分点。纤维素酶和木聚糖酶混合辅助水蒸气蒸馏法提取柑橘属精油是一种高效的精油提取方法,为橘皮废料的无害化处理提供了一种解决方案。
In this paper,the essential oil of orange peel was extracted by steam distillation from summer orange by enzymolysis of cellulase and xylanase.The optimum enzymolysis temperature,enzymolysis pH and enzymolysis time of mixed enzymes were investigated by single factor experiment.Based on the results of single factor test,the optimum extraction conditions of essential oil from orange peel were investigated by response surface method.The results showed that the optimum addition amount of cellulase was 1.2%by weight of orange peel,and the optimum addition amount of xylanase was 2.2%by weight of orange peel.The optimum conditions of mixed enzyme assisted steam distillation for extraction of orange peel essential oil were as follows:the enzymatic hydrolysis temperature was 41℃,the enzymatic hydrolysis pH was 6.337,and the enzymatic hydrolysis time was 1.4 h.under above conditions,the extraction yield of orange peel essential oil was 1.54%,which was higher than that without adding enzyme by 1.02 percentage point.Extraction of citrus essential oil by steam distillation assisted by cellulase and xylanase is an efficient method,which provides a solution for the harmless treatment of orange peel waste.
