毛霉菌菌丝体多糖的提取工艺优化和抗氧化活性 被引量:1

Optimization of Extraction Technology and Antioxidant Activity of Polysaccharide from Mucor Sp.Mycelia

摘要 优化毛霉菌菌丝体多糖(MPS)的提取条件,研究其抗氧化活性。利用单因素试验和正交试验考察料液比、提取时间、提取温度和提取次数对菌丝体多糖提取率的影响,检测MPS对DPPH、ABTS和羟自由基的清除活性评价其抗氧化活性,研究MPS对人肝癌HepG2细胞增殖的影响。毛霉菌菌丝体多糖的最佳提取条件为料液比1∶50 (g/mL),提取时间2 h,提取温度为80℃,重复提取2次;在此条件下, MPS的提取率为18.24%, MPS能够清除DPPH、ABTS和羟自由基,在5 mg/mL时对三种自由基的清除率分别是42.76%, 40.59%和42.76%。同时, MPS对人肝癌Hep G2细胞的增殖无显著影响。文章确定了毛霉菌菌丝体多糖的最佳提取工艺,并发现该多糖具有一定的抗氧化活性。 Optimize the extraction method of polysaccharides from Mucor sp.mycelia,and study its antioxidant activity.The effects of solid/liquid ratio,extraction time,extraction temperature and extraction times on the extraction rate of polysaccharides were investigated by single factor test and orthogonal test.The antioxidant activity was evaluated by the scavenging activity of polysaccharide on DPPH,ABTS and hydroxyl radicals,and we studied the effect of polysaccharide on the proliferation of HepG2 cells.The optimum condition was material/liquid ratio of 1:50(g/mL),extraction time for 2 h,temperature at 80℃,and repeated extraction twice.The extraction rate of polysaccharide was 18.24%under this condition.MPS could scavenge DPPH,ABTS and hydroxyl radicals,and the scavenging rates of free radicals at 5 mg/mL were 42.76%,40.59%and 42.76%,respectively.Meanwhile,MPS had no significant effect on the proliferation of HepG2 cells.In this paper,the optimal extraction condition of polysaccharide was determined,and it was found that the polysaccharide had certain antioxidant activity.
