摘要
为了探究枳壳黄酮提取物的抗肿瘤活性,以重庆枳壳为研究对象,通过超高效液相色谱法测定不同枳壳中的新橙皮苷、柚皮苷、橙皮苷和芸香柚皮苷的含量。使用超声波提取法提取枳壳黄酮,结合methyl thiazolyl blue tetrazolium bromide (MTT)比色法和EGFP-Annexin V/PI染色法分析其抗肿瘤活性。结果发现,不同枳壳样品的柚皮苷、新橙皮苷、橙皮苷和芸香柚皮苷的含量存在较大的差异,其中柚皮苷含量在0.23%~10.78%,新橙皮苷含量在0.17%~7.17%。22个样品中符合2020年版《中国药典》标准的有18个样品,合格率为81.8%。超声波辅助提取枳壳黄酮提取物的提取率为11.82%,与阴性对照相比,枳壳黄酮提取物对HCT116结直肠癌细胞具有显著抑制效果(P<0.05)且呈量效关系, 48 h内黄酮提取物(400μg/mL)对结直肠癌肿瘤细胞的抑制率为60.83%, IC50值为317.2μg/mL,细胞晚期凋亡率为13.7%。枳壳黄酮提取物能够抑制结直肠癌肿瘤细胞的增殖活性,为枳壳黄酮抗肿瘤活性的研究提供理论依据。
In order to explore the antitumor activity of Citrus flavonoids extracts,taking Auranti Fructus from Chongqing as the research object,the neohesperidin,naringin,hesperidin,and narirutin in different Citrus were determined by ultra-high performance liquid chromatography.The flavonoids of Auranti Fructus were extracted by ultrasonic extraction,and their antitumor activity was analyzed by MTT and EGFP-Annexin V/PI staining.The results showed that the contents of neohesperidin,naringin,hesperidin and narirutin in different Auranti Fructus were quite diferent.Among them,the content of naringin ranged from 0.23%to 10.81%,and the content of neohesperidin ranged from 0.17%to 7.17%.Among the 22 samples,18 samples were in conformity with the standard of Chinese Pharmacopoeia(2020),with a qualification rate of 81.8%.The extraction rate of flavonoids extracted by ultrasonic wave was 11.82%.Compared with the negative control,aurantium flavonoids had a significant inhibitory effect on HCT116 colorectal cancer cells(P<0.05)and showed a doseeffect relationship.The inhibitory rate of flavonoid extract(400μg/mL)on colorectal cancer tumor cells within 48 hours was 60.83%,ICso value was 317.2μg/mL,and the late apoptosis rate was 13.7%.The flavonoid extract from Auranti Fructus can inhibit the proliferative activity of colorectal cancer cells,providing a theoretical basis for the study of the antitumor activity of AurantiiFructusflavone.
