颐脑解郁方对脑出血后抑郁大鼠神经再生和修复的影响及机制 被引量:1

Effect and mechanism of Yinao Jieyu Decoction on nerve regeneration and repair in rats with depression after cerebral hemorrhage

摘要 目的 探讨颐脑解郁方对脑出血后抑郁大鼠神经再生和修复的影响及机制。方法 将大鼠随机分为空白组、假手术组、模型组、氟西汀组、颐脑解郁方组,各8只。模型组、氟西汀组、颐脑解郁方组采用尾壳核注射胶原酶诱发脑出血联合慢性不可预知温和刺激(CUMS)建立抑郁模型。在CUMS开始首日同时给予药物灌胃,氟西汀组给予盐酸氟西汀2.33 mg/(kg·d),颐脑解郁方组给予颐脑解郁方9.92 g/(kg·d),空白组、假手术组、模型组均给予蒸馏水10 mL/kg,灌胃4周,分别于造模前及造模第1、2、4周测量体质量、行蔗糖水试验,并于第4周进行OFT旷场实验,Western blotting法检测海马组织中Nogo-A、NgR、RhoA蛋白表达。结果 与空白组、假手术组比较,模型组体质量、蔗糖水消耗量、直立次数少(P<0.05);与模型组比较,氟西汀组和颐脑解郁方组以上指标均高(P<0.05)。与空白组、假手术组比较,模型组海马组织中Nogo-A、NgR、RhoA蛋白表达均高(P<0.05);与模型组比较,氟西汀组、颐脑解郁方组海马组织中Nogo-A、NgR、RhoA蛋白表达低(P<0.05);颐脑解郁方组海马组织中Nogo-A蛋白表达低于氟西汀组(P<0.05),NgR蛋白表达高于氟西汀组(P<0.05),2组海马组织中RhoA蛋白表达比较差异无统计学意义(P>0.05)。结论 颐脑解郁方可通过下调轴突生长抑制因子Nogo-A、NgR、RhoA蛋白表达,促进脑出血后抑郁大鼠的神经再生和修复。 Objective To explore the effect and mechanism of Yinao Jieyu Recipe on nerve regeneration and repair in depressed rats after cerebral hemorrhage.Methods All rats were randomly divided into sham group,sham-operating group,model group,Fluoxetine group,and Yinao Jieyu Decoction group with 8 in each group.The model group,fluoxetine group and Yinao Jieyu recipe group was established depression models by injecting collagenase into caudate putamen to induce cerebral hemorrhage combined with chronic unpredictable mild stimulation(CUMS).On the first day of CUMS,drugs were given to the stomach at the same time.The fluoxetine group was given fluoxetine hydrochloride 2.33 mg/(kg·d),the Yinao Jieyu recipe group was given Yinao Jieyu recipe 9.92 g/(kg·d),and the blank group,sham operation group and model group were given distilled water 10 mL/kg,which was given to the stomach for 4 weeks,body weight and sucrose water test were measured before modeling and at the 1st,2nd and 4th week,respectively,and the OFT open field experiment was conducted at the 4th week.The expression of Nogo-A,NgR and RhoA protein in hippocampus was detected by Western blotting.Results Compared with the blank group and sham operation group,the body weight,sucrose water consumption and the times of standing upright in the model group were less(P0.05).Compared with the model group,the above indexes in fluoxetine group and Yinao Jieyu recipe group were higher(P0.05).Compared with the blank group and sham operation group,the expression of Nogo-A,NgR and RhoA protein in hippocampus of model group was higher(P0.05).Compared with the model group,the expressions of Nogo-A,NgR and RhoA in hippocampus of fluoxetine group and Yinao Jieyu recipe group were lower(P0.05).The expression of Nogo-A protein in hippocampus of Yinao Jieyu recipe group was lower than that of fluoxetine group(P0.05),and the expression of NgR protein was higher than that of fluoxetine group(P0.05).There was no significant difference in the expression of RhoA protein between the two groups(P0.05).Conclusion Yinao Jieyu Recipe can promote nerve regeneration and repair in depressed rats after cerebral hemorrhage by down-regulating the expression of axon growth inhibitory factors Nogo-A,NgR and RhoA.
