杜仲组织培养外植体的选择和消毒

Selection and Disinfection of Explants for Tissue Culture of Eucommia ulmoides

[目的]找出杜仲外植体选择和消毒的适宜方式方法。[方法]以华中8号杜仲优良母株为外植体材料,进行杜仲的植物组织培养。在进行杜仲组培外植体选取和处理时,研究选取不同部位、采用不同消毒剂对杜仲离体组培的影响。[结果]进行杜仲组织培养外植体选取时,4—5月所选取的顶芽为最适合的材料;预处理后放入流水冲洗2.0~2.5 h,先用酒精浸泡30~40 s,再用0.1%HgCl2浸泡消毒8 min,消毒效果最好,存活率最高。[结论]该研究为杜仲的组培快繁技术提供了依据。

[Objective]The aim of this study was to find out the suitable method for explant selection and disinfection of Eucommia ulmoides.[Method]With the excellent mother plant of Huazhong 8 E.ulmoides as the explant material,the plant tissue culture of E.ulmoides was carried out.In the selection and treatment of E.ulmoides tissue culture explants,the effects of different parts and different disinfectants on the in vitro tissue culture of E.ulmoides were studied.[Result]When selecting explants for tissue culture of E.ulmoides,the apical buds selected from April to May were the most suitable materials.After pretreatment,it was rinsed with running water for 2.0-2.5 hours,soaked in alcohol for 30-40 seconds,and then soaked in 0.1%HgCl2 for 8 minutes.The disinfection effect was the best and the survival rate was the highest.[Conclusion]The research provided basis for tissue culture and rapid propagation technology of E.ulmoides.
