摘要
亚胺是有机合成中一类性质活泼的重要反应单元,可发生加成、还原等一系列化学反应,生成广泛存在于天然产物和活性分子中的含氮化合物。同时,亚胺化合物还具有抗菌等独特的生物活性和药理活性,而亚胺因其特定的结构容易水解,在硅胶板上分解成原料,难以通过TLC观测反应进程,且分离纯化损失大。因此,如何高效构建不需要进一步纯化的亚胺化合物成为了当下有机合成中的难点。本研究发展了一种新型的亚胺合成方法,使用氘代氯仿作为反应溶剂,无水硫酸镁为吸水剂,在90~130℃下回流分水,核磁共振仪监测反应进程,反应时间最快2小时,产率最高达到98%。该方法操作便捷,原子经济性高,环境友好,所使用反应溶剂可以回收使用。
Imines are a kind of active and important reaction units in organic synthesis.A series of chemical reactions such as addition and reduction can occur to produce nitrogen-containing compounds widely existing in natural products and active molecules.At the same time,imine compounds also have unique biological and pharmacological activities such as antibacterial.However,imine compounds are easy to hydrolysis due to its specific structure and decompose into raw materials on silica gel plate.It is difficult to observe the reaction process by TLC,and the separation and purification loss is great.Therefore,how to efficiently construct imine compounds without further purification has become a difficulty in organic synthesis.In this study,a novel imine synthesis method was developed.Deuterium chloroform was used as the reaction solvent and anhydrous magnesium sulfate was used as the water absorbent.The reflux water was separated at 90℃ to 130℃,and the reaction process was monitored by NMR.The method is convenient,economical and environmentally friendly,and the solvent used can be recycled.
