摘要
研究了由五种中草药组成的复合提取液的制备,对其进行了总黄酮含量测定,并通过DPPH自由基清除测试、细胞I型前胶原含量测试、人体功效测试等验证了复合提取物的抗氧化、紧致抗皱功效。结果显示,经乙醇水溶液超声提取、AB-8柱层析吸附制得的复合提取液,其平均总黄酮含量高达5.96 mg/mL,RSD值为1.45%。复合提取液对DPPH自由基抑制率IC50值为45.30μg/mL;在细胞测试中,0.2%浓度时,胶原蛋白上调率为88.34%(P<0.001);在人体功效测试中,20名志愿者使用含5%复合提取液的产品28天后,结果显示,皱纹分值均值在D14、D28天分别降低了18.87%、24.48%(P<0.05),皮肤紧致度F4均值在D14、D28分别降低了6.84%、23.62%(P<0.01)。综上所述,该复合提取液有良好的抗氧化、紧致和抗皱功效,在化妆品中有较好的应用前景。
The preparation of the composite extract composed of five Chinese herbal medicines was studied,and the content of total flavonoids was determined.The effect of antioxidant,firming and anti-wrinkle were studied by several methods,such as DPPH radical scavenging experiment,cellular procollagen type I content test,and in vivo study.In this paper,the composite ingredients were extracted by ultrasonic tectnology with aqueous ethanol solution and adsorbed by the AB-8 column chromatography.The results showed that the average total flavonoid content of the composite extract was up to 5.96 mg/mL,and the relative standard deviation(RSD)was 1.45%.As for DPPH radical scavenging experiment,the IC50 of this ingredient was 45.30μg/mL.In cell tests,at the concentration of 0.2%,the up-reguation rate of collagen was 88.34%(P<0.001).For in vivo efficacy performance,20 volunteers used the product containing 5%of the extract for 28 days,the results indicated that the average value of wrinkle score decreased by 18.87% and 24.48% on D14 and D28,respectively(P<0.05),and the average value of skin firmness F4 decreased by 6.84%and 23.62%on D14 and D28,respectively(P<0.01).These findings suggest that the composite extract has good antioxidant,firming and anti-wrinkle effect,which with great prospect in the application of cosmetics.
