摘要
目的:基于网络药理学方法探索黄芪三七配伍治疗肺纤维化的机制研究。方法:基于TCMSP、Pubchem、SwissTarget Predicition、GeneCards、OMIM、TTD、DisGeNET等数据库,筛选出药物以及疾病的靶点基因,使用venny 2.1在线软件作图工具平台取其交集,Cytoscape3.9.0创建其药物-有效成分-基因调控网络,借助String数据库建立其蛋白互作PPI网络进行核心基因的筛选,使用DAVID数据库进行GO和KEGG富集分析,通过微生信进行可视化分析。结果:黄芪三七有效成分共31个,药物疾病交集靶点基因204个,作用于疾病的核心基因有KT1、IL-6、VEGFA、EGFR、JUN、LYN、CASP3、HMOX1等。KEGG信号通路159个,包括Pathways in cancer、EGFR tyrosine kinase inhibitor resistance、PI3K-Akt signaling pathway、HIF-1 signaling pathway、Proteoglycans in cancer、Rap1 signaling pathway、Ras signaling pathway等。结论:黄芪三七药物配伍在抗纤维化过程中可能通过多成分、多靶点、多通路协调整合发挥作用,为后续动物实验提供有效参考。
Objective:To explore the mechanism of Astragalus panax notoginseng in the treatment of pulmonary fibrosis based on network pharmacology method.Methods:Target genes of drugs and diseases were screened out based on databases such as TCMSP,Pubchem,Swiss Target Predicition,GeneCards,OMIM,TTD,DisGeNET,etc.Venny 2.1 online software mapping tool platform was used to obtain their intersection.Cytoscape3.9.0 is used to create the drug-active ingredient-gene regulatory network,and String database is used to establish the protein interaction PPI network to screen the core genes,DAVID database is used for GO and KEGG enrichment analysis,and microbiosis is used for visual analysis.Results:There were 31 active components of Astragalus and Notoginseng,204 target genes of drug diseases.The core genes acting on diseases included KT1,IL-6,VEGFA,EGFR,JUN,LYN,CASP3,HMOX1,etc.159 KEGG signaling pathways,These include Pathways in cancer,EGFR Tyrosine kinase inhibitor resistance,PI3K-AKT signaling pathway,and HIF-1 signaling Pathway,Proteoglycans in cancer,Rap1 Signaling Pathway,Ras Signaling Pathway,etc.Conclusion:Astragalus-Panax notoginseng compatibility can exert anti-PF effect through multi-component,multi-target and multi-pathway coordination and integration,which provides reference for subsequent cell and animal experiments.
