炙甘草汤调节免疫细胞自噬干预房颤的生物信息机制探讨 被引量:1

Study on the Bioinformatics Mechanism of Zhigancao Decoction in Regulating Immune Cell Autophagy and Intervening Atrial Fibrillation

摘要 目的运用生物信息学方法预测炙甘草汤调节免疫细胞自噬干预房颤(Atrial fibrillation,AF)的关键成分和潜在靶点,进而探讨其治疗AF的作用机制。方法分析基因表达数据库(GEO)中AF相较于窦性心律患者差异表达的自噬基因,计算22种免疫细胞浸润程度。LASSO回归筛选关键靶点并分析与免疫细胞的相关性,并与炙甘草汤相关靶点取交集后建立“炙甘草汤-活性成分-AF中免疫细胞自噬相关靶点”网络,随后进行蛋白相互作用、基因本体(GO)功能富集与京都基因与基因组百科全书(KEGG)通路富集分析。结果95个差异表达自噬基因中13个基因可作为AF的诊断标记,肥大细胞是AF与窦性心律心肌组织内差异浸润的免疫细胞,与ATG4C等自噬基因有密切关系。炙甘草汤作用于42个AF中免疫细胞自噬相关靶点,核心靶标包括白细胞介素-1β(IL-1β)、细裂原活化蛋白激酶1(MAPK1)和T细胞受体信号传导通路等关键通路。结论炙甘草汤可能通过调节免疫细胞自噬和炎症反应而达到治疗AF的目的。 Objective To predict the key components and potential targets of Zhigancao decoction in regulating immune cell autophagy in intervention of atrial fibrillation(AF)by bioinformatics method,and to explore the mechanism of Zhigancao decoction in treating AF.Methods the autophagy genes differentially expressed in AF and sinus rhythm patients in GEO database were analyzed,and the infiltration degree of 22 kinds of immune cells was calculated.LASSO regression was used to screen the key targets and analyze the correlation with immune cells,and the network of“Zhigancao decoction-active ingredient-immune cell autophagy-related targets in AF”was established after intersecting with Zhigancao decoction.Then protein interaction,GO functional enrichment and KEGG pathway enrichment were analyzed.Results 13 of the 95 differentially expressed autophagy genes could be used as diagnostic markers for AF.Mast cells were differential infiltrating immune cells in the myocardium of AF and sinus rhythm,and were closely related to autophagy genes such as ATG4C.Zhigancao decoction acts on 42 autophagy-related targets of immune cells in AF,including key pathways such as IL1B,MAPK1 and T cell receptor signal transduction pathway.Conclusion Zhigancao decoction may achieve the purpose of treating AF by regulating immune cell autophagy and inflammation.
