核黄素营养不良对大鼠肠道菌群的影响

EFFECTS OF RIBOFLAVIN MALNUTRITION ON GUT MICROBIOTA IN RATS

摘要 目的 探究不同核黄素营养水平对大鼠肠道菌群的影响。方法 将36只雄性Wistar大鼠按体重随机分为正常对照组(normal control,NC),核黄素缺乏组(riboflavin deficiency group,RD),核黄素不足组(riboflavin insufficiency group,RI)和核黄素缺乏对喂组(riboflavin deficiency pair-fed group,PF),分别喂养核黄素含量6mg/kg、<1mg/kg、3mg/kg和6mg/kg的AIN-93合成饲料3个月,实验结束时取血清、结肠和肠道内容物,分别测定核黄素营养状况指标、肠黏膜结构与紧密连接蛋白表达、短链脂肪酸和肠道菌群变化。结果 与NC组相比,核黄素营养不良大鼠血清及肝脏中核黄素及其衍生物含量下降,血清谷胱甘肽还原酶活性下降,肠黏膜结构发生损伤,紧密连接蛋白ZO-1、Claudin-1表达量降低,短链脂肪酸含量显著降低,肠道菌群多样性降低。RD组中Prevotella_9、Alloprevotella、Phascolarctobacterium、Holdemania、Parasutterella的相对丰度显著增加(P<0.05),RD和RI组中[Eubacterium]_fissicatena_group的相对丰度显著减少(P<0.05),PF组中Prevotella_9、Alloprevotella、Phascolarctobacterium、Parasutterella的相对丰度显著增加(P<0.05)。结论 核黄素营养不良对大鼠肠道微生物菌群丰度及结构具有显著影响,导致短链脂肪酸产生减少、肠黏膜屏障功能下降。[营养学报,2023,45(2):163-171] Objective This study was aimed to investigate the effects of riboflavin status on gut microbiota in rats.Methods 36 male Wistar rats were randomly divided into four groups based on body weight:normal control group(NC),riboflavin deficiency group(RD),riboflavin insufficiency group(RI)and riboflavin deficiency pair-fed group(PF).They were fed AIN-93 diets containing 6mg/kg,<1mg/kg,3mg/kg and 6mg/kg riboflavin for 3 month,respectively.The riboflavin status,histomorphology of colonic tissues,short-chain fatty acids(SCFAs)in colonic contents,expression of tight junction proteins,and gut microbiota in colonic contents were assayed.Results Compared to the NC group,the levels of riboflavin and its derivatives in the serum and liver,the activity of glutathione reductase in the serum,the expressions of ZO-1 and Claudin-1,the contents of acetic acid,propionic acid,butyric acid,valeric acid,isobutyric acid and isovaleric acid were significantly decreased in the riboflavin-deficient rats.The diversity of intestinal flora was decreased.Meanwhile,histomorphology of colonic tissues was significantly damaged in response to riboflavin deficiency.In comparison with the NC group,the relative abundance of Prevotella_9,Alloprevotella,Phascolarctobacterium,Holdemania,and Parasutterella were significantly increased in the RD group,[Eubacterium]_fissicatena_group was significantly decreased in the RD and RI groups,and Prevotella_9,Alloprevotella,Phascolarctobacterium,and Parasutterella were significantly increased in the PF group.Conclusion Riboflavin malnutrition has a significant impact on the diversity and composition of gut microbiota,and leads to decline of SCFAs and disruption of intestinal mucosal barrier function.[ACTA NUTRIMENTA SINICA,2023,45(2):163-171]
