不同生境金钗石斛伴生苔藓内生真菌群落结构多样性及其分布规律 被引量:2

Community and diversity of endophytic fungi associated with concomitant moss of Dendrobium nobile in different habitats

摘要 目的:通过对不同生境金钗石斛伴生苔藓内生真菌群落组成和多样性的研究,寻找野生与大棚生境的差异真菌,为金钗石斛人工栽培提供参考依据。方法:利用lIllumina HiSeq高通量测序技术对野生和大棚生境中金钗石斛伴生苔藓内生真菌群落组成与多样性进行分析,采用FUNGuild分析真菌群落功能。结果:不同生境金钗石斛伴生苔藓内生真菌多样性与菌群结构存在差异。野生丹霞石生境金钗石斛伴生苔藓内生菌多样性显著高于大棚生境。门水平上,子囊菌门(Ascomycota)和担子菌门(Basidiomycota)为金钗石斛伴生苔藓内生真菌的优势菌门。在属水平上不同生境伴生苔藓内生真菌优势菌属差异较大,如野生丹霞石生境丰度前3位优势菌属为Apiotrichum、枝孢属Cladosporium、Conlarium,大棚丹霞石生境相对丰度前3位的优势菌属是Apiotrichum、黑孢霉属Nigrospora、链格孢属Alternaria。野生生境中Cladosporium、Conlarium、枝孢瓶霉属Cladophialophora和Diutina的相对丰度显著高于3种大棚生境的苔藓样品。此外,4个生境伴生苔藓内生真菌均存在特有属,如野生丹霞石生境中特有属相对丰度的前3位是Conlarium、Auricularia和胶鞘盘菌属Pezoloma;大棚丹霞石生境中特有属相对丰度的前3位是柱霉属Scytalidium、柱锈耳属Eocronartium和嗜热子囊菌属Thermoascus。结论:本研究为进一步开发利用金钗石斛伴生苔藓内生真菌资源,研究金钗石斛与伴生苔藓互利共生机制提供参考依据。 Objective:To analyze the difference of community and structure of endophytic fungi associated with concomitant moss of Dendrobium nobile and provided the reference for the artificial cultivation of Dendrobium nobile.Methods:Illumina HiSeq high-throughput sequencing technology and bioinformatics tools were used to analyze the difference of the community composition and diversity of endophytic fungi associated with concomitant moss of Dendrobium nobile in wild and greenhouse habitats.FUNGuild was used to analyze the functions of fungal communities.Results:There was different of the diversity and community of endophytic fungi associated with mosses of Dendrobium nobile from different habitats.The diversity of endophytic fungi in the wild habitat was significantly higher than that in the greenhouse habitat.At the level of phylum,Ascomycota and Basidiomycota were the dominant endophytic fungi.At the genus level,the dominant genera of endophytic fungi varied in different habitats.The top three dominant genera of relative abundance were Apiotrichum,Cladosporium and Conlarium in Wild Danxia habitat,while the top three dominant genera of relative abundance are Apiotrichum,Nigerospora and Alternaria in the greenhouse Danxia Stone habitat.The relative abundance of Cladosporium,Conlarium,Cladosporium in wild habitat was significantly higher than those in greenhouse habitat.Additionally,the endophytic fungi in the four habitats all have their own specific genera.For example,Conlarium,Auricularia and Pezoloma were the top 3 specific genera in the wild Danxia rock habitat;Scytalidium,Eocronartium and Thermoascus were the top 3 specific genera in the greenhouse Danxia habitat.Conclusion:This study provides a reference for further development and utilization of the endophytic fungi resources of Dendrobium nobile concomitant moss,as well as the research on mechanism of symbiosis between Dendrobium nobile and concomitant moss.
