蓬莪术醋制入肝“毒-效”关系对比研究

摘要 目的:探究蓬莪术醋制入肝后的“毒-效”关系变化。方法:将小鼠分为7组:对照组,蓬莪术生品及醋品挥发油高、中、低剂量组(1.64、0.82、0.41 g/kg),每组10只,腹腔注射给药(25 mL/kg),观察14 d,记录其急性毒性的生理反应、脏器指数及肝组织HE染色情况;将大鼠分为6组:对照组,模型组,蓬莪术生品及醋品高、低剂量组(2.0、1.0 g/kg),除对照组外,各组以皮下注射3 mL/kg 50%CCl_4橄榄油溶液建立大鼠肝纤维化模型,灌胃给药(10 mL/kg),1次/d,常规饲养7周后取血清,测定白蛋白(ALB)、谷丙转氨酶(ALT)、谷草转氨酶(AST)及总胆红素(TBIL)水平,取肝脏拍照并做HE、天狼猩红、Masson染色处理。结果:蓬莪术挥发油显著升高小鼠肝、脾的脏器指数(P<0.05),且醋制后的挥发油能升高小鼠LD_(50)值,改善肝组织的病理变化;此外,蓬莪术醋制后可改善肝脏受损、纤维胶原沉积,降低ALT(P<0.05)水平,保肝降酶以延缓肝纤维化病程,其中醋莪术低剂量组疗效最佳。结论:蓬莪术醋制后可增强对肝脏的保护作用,缓解峻猛药性的同时,还能增强对肝纤维化大鼠的疗效。 Objective:To study the changes of‘toxicity-efficacy’relationship after vinegar-processing Curcuma phaeocaulis VaL.via leading drug into liver.Methods:The mice were divided into 7 groups:control group,high-dose Rolatile oil group,medium-dose volatile oil group and low-dose volatile oil group(1.64,0.82,0.41 g/kg),10 mice in each group.The mice were given intraperitoneal injection(25 mL/kg)for 14 days,and the physiological reaction of acute toxicity,organ index and liver tissue HE staining were recorded.Besides,the rats were divided into 6 groups:control group,model group,and high-dose group and low-dose group(2.0,1.0 g/kg).Except for the control group,the liver fibrosis model of rats in each group was established by subcutaneous injection with 50%CCl4 olive oil solution,which was given by gavage(10 mL/kg)once a day,and the rats were fed routinely for 7 weeks.Serum samples were collected from abdominal aorta of rats to determine the levels of ALB,ALT,AST and TBIL.After death,the liver was taken and photographed and treated with HE,Sirius scarlet and Masson staining.Results:The volatile oil of Curcuma phaeocaulis VaL.significantly increased the organ index of liver and spleen of mice(P<0.05),and the volatile oil after vinegar-processing could increase the LD50 value of mice,and improve the pathological changes of liver tissue.In addition,after vinegar-processing,Curcuma phaeocaulis VaL.could improve liver damage,fibrous collagen deposition,reduce the levels of ALT(P<0.01)to delay the course of liver fibrosis,and the low-dose group had the best effect.Conclusion:After vinegar-processing,Curcuma phaeocaulis VaL.can enhance the protective effect on liver,reduce toxicity,and enhance the curative effect on liver fibrosis in rats.
