含共轭烯炔键的不饱和芳香酯的合成

摘要 含有共轭烯炔键的化合物可基于高反应性的不饱和键进行多样化衍生,不仅能与过渡金属配位对不饱和键进行官能团修饰,还能通过不饱和键与药物分子结合后进行药品改性。因此,含有共轭烯炔键的化合物在天然产物合成和药物分子合成中应用广泛。制备了磷叶立德,并通过维蒂希(Wittig)反应、羰基还原、羟基酯化等,合成了一类含共轭烯炔键的不饱和芳香酯,6步总产率在15%~35%。该类化合物的成功合成,极大拓展了不饱和芳香酯的结构类型,并可作为重要的小分子砌块应用于天然产物或药物分子合成领域。 Compounds containing conjugated alkyne bonds can be diversified based on highly reactive unsaturated bonds.They can not only coordinate with transition metals to modify unsaturated bonds with functional groups,but also modify drugs by combining unsaturated bonds with drug molecules.Therefore,it is widely used in the synthesis of natural products and drug molecules.Phosphorus ylide was prepared,and a class of unsaturated aromatic esters containing conjugated alkyne bonds were synthesized through Wittig reaction,carbonyl reduction,hydroxyl esterification,etc.,with a total yield of six steps was 15%-35%.The successful synthesis of these compounds greatly expands the structural types of unsaturated aromatic esters and can be used as important small molecule blocks in the synthesis of natural products or drug molecules.
