摘要
该研究通过透明圈法、淀粉酶活力测定、分子生物学鉴定及粗酶液的温度耐受性分析,从浓香型白酒窖泥中分离筛选优良的高产淀粉酶菌株,并对其进行常压室温等离子体(ARTP)诱变选育,最后研究其对纤维素及半纤维素的分解能力。结果表明,筛选得到一株高产淀粉酶活力的优良菌株TS-5,经鉴定为厚壁类芽孢杆菌(Paenibacillus cineris),其所产淀粉酶活力为55.23 U/m L,可耐受70℃的高温处理。通过ARTP诱变得到高产淀粉酶活力的突变菌株TS-5-ARTP,其淀粉酶活力(95.65 U/m L)为出发菌株的1.73倍,该突变菌株具有良好的分解利用纤维素及半纤维素的能力。
High amylase-producing strain was isolated and screened from the pit mud of strong-flavor(Nongxiangxing)Baijiu through transparent circle method,amylase activity determination,molecular biology identification,and temperature tolerance analysis of crude enzyme solution,excellent amylase-producing strains were isolated and screened from the pit mud of strong-flavor Baijiu and then induced by atmospheric pressure and room temperature plasma(ARTP),and the decomposition ability on cellulose and hemicellulose was studied.The results showed that strain TS-5 with high amylase activity was screened and obtained,which was identified as Paenibacillus cineris.The amylase activity was 55.23 U/ml,which could tolerant 70℃high temperature treatment.The mutant strain TS-5-ARTP with high amylase activity was obtained by ARTP mutagenesis,and its amylase activity was 1.73 times as high as that of the original strain.The strain had good ability to decompose and utilize cellulose and hemicellulose.
