厚朴排气合剂对便秘大鼠肠道分泌功能及肠道菌群的影响

Effects of Houpo Paiqi Mixture on intestinal secretion function and gutmicrobiota in constipated rats

目的:观察厚朴排气合剂对便秘大鼠肠道分泌和肠道菌群的影响。方法:将42只SD大鼠随机分为对照组、模型组和中药组,每组14只。模型组及中药组给予盐酸洛哌丁胺8 mg/(kg·d)灌胃,造模成功后中药组给予10.5 mL/(kg·d)的厚朴排气合剂灌胃治疗1周。通过粪便含水量评价厚朴排气合剂治疗效果,通过黏膜短路电流测定肠道分泌的变化,通过16S rRNA高通量测序分析肠道菌群的变化,通过液相色谱-串联质谱技术测定短链脂肪酸含量的变化。结果:与对照组比较,模型组大鼠的粪便含水量和结肠黏膜短路电流显著下降(P<0.05);与模型组比较,中药组大鼠的粪便含水量和结肠黏膜短路电流显著上升(P<0.05)。与对照组比较,模型组大鼠的瘤胃球菌属丰度明显下降,普氏菌属、乳杆菌属的相对丰度明显上升,总短链脂肪酸含量、乙酸和丙酸含量显著下降(P<0.05);与模型组比较,中药组大鼠的普氏菌属、乳杆菌属的相对丰度明显下降,拟杆菌属、阿克曼菌属、布劳特氏菌属的相对丰度明显上升,乙酸含量显著升高(P<0.05),异戊酸和己酸含量显著下降(P<0.05)。结论:厚朴排气合剂可能通过促进肠道分泌、调整肠道菌群结构及短链脂肪酸的含量,从而改善大鼠的便秘症状。

Objective To observe the effects of Houpo Paiqi Mixture on intestinal secretion and gut microbiota in constipated rats.Methods Forty-two SD rats were randomly divided into control group(NC),model group(CM)and Traditional Chinese Medicine group(HH),with 14 rats in each group.CM group and HH group were given loperamide hydrochloride eight mg/(kg·d),and the HH group was given 10.5 mL/(kg·d)of Houpo Paiqi Mixture intragastric treatment for one week after successful modeling.The therapeutic effect of Houpo Paiqi Mixture was evaluated by fecal water content,the changes of intestinal secretion function were determined by mucosal short-circuit current,the changes of intestinal flora were analyzed by 16S rRNA high-throughput sequencing,and the changes of short-chain fatty acid content were determined by liquid chromatoc-tandem mass spectrometry.Results Compared with NC group,fecal water content and short circuit current of colonic mucosa in model group were significantly decreased(P<0.05).Compared with CM group,fecal water content and short circuit current of colonic mucosa of rats in HH group was significantly increased(P<0.05).Compared with NC group,the abundance of Ruminococcus in CM group was significantly decreased,the relative abundance of Prevotella and Lactobacillus was significantly increased,and the contents of total short-chain fatty acids,acetic acid,propionic acid were significantly decreased(P<0.05).Compared with CM group,the relative abundance of Prevotella and Lactobacillus in HH group were significantly decreased,while Bacteroides,Akkermansia and Blautia were significantly increased,and the acetic acid content was significantly increased(P<0.05),isovaleric acid and caproic acid contents were significantly decreased(P<0.05).Conclusion Houpo Paiqi Mixture may improve constipation symptoms in rats by promoting intestinal secretion,adjusting intestinal flora structure and short-chain fatty acid content.
