商陆不同组分对上焦水饮内停大鼠模型药效学研究

Pharmacodynamic Study of Different Components of PHYTOLACCAE RADIX on the Shangjiaoshuiyinneiting(上焦水饮内停)Model Rats

摘要 目的:探讨商陆水煎液及不同组分对大鼠上焦水饮内停模型药效学研究,寻找商陆对上焦水饮内停模型作用的药效物质基础。方法:采用复合造模因素(大鼠腹部皮下注射异丙肾上腺素+气管插管+寒冷刺激)建立上焦水饮内停模型,模型建立后开始给予商陆水煎液0.81 g/kg、多糖组分0.31 g/kg、皂苷组分0.22 g/kg、酚酸组分0.17 g/kg、鞣质组分0.09 g/kg,连续灌胃给药14 d。试验结束后检测大鼠血清白蛋白(ALB)、肌酸激酶(CK)含量,肺干湿比,肺泡灌洗液中蛋白含量,心、肺脏指数,观察心、肺组织病理切片,综合评价商陆水煎液及不同组分对上焦水饮内停模型大鼠利水功效。结果:与正常对照组比较,模型对照组血清ALB含量显著降低(P<0.01),肺泡灌洗液中蛋白浓度、肺干湿比、CK活力以及心脏、肺脏指数明显升高(P<0.05或P<0.01),心、肺组织受损较严重,上焦水饮内停模型建立成功;与模型对照组比较,商陆水煎液0.81 g/kg组可明显升高血清ALB(P<0.05),明显降低CK活力、肺泡灌洗液中蛋白含量、心脏指数、肺干湿比(P<0.05或P<0.01);多糖组分0.31 g/kg组可明显升高血清ALB(P<0.05),明显降低CK活力、心脏指数、肺脏指数、肺干湿比(P<0.05或P<0.01);皂苷组分0.22 g/kg组明显升高血清ALB含量(P<0.05),明显降低CK活力(P<0.05);商陆水煎液0.81 g/kg组、多糖组分0.31 g/kg组对上焦水饮内停大鼠心、肺病理状况有明显改善作用。结论:商陆水煎液可改善上焦水饮内停模型大鼠的心肺损伤。多糖组分和皂苷组分对上焦水饮内停模型大鼠具有不同程度的治疗作用,其中多糖组分治疗作用明显。 Objective:To study the pharmacodynamics of PHYTOLACCAE RADIX decoction and its components on the Shangjiaoshuiyinneiting(上焦水饮内停)model in rats and explore the pharmacodynamic material basis.Methods:The Shangjiaoshuiyinneiting model was established by a multi-factor compound method(abdominal subcutaneous injection of isopropyl adrenaline+endotracheal intubation+cold stimulation).After modeling,rats were orally administered with PHYTOLACCAE RADIX decoction(0.81 g/kg),polysaccharide extract(0.31 g/kg),saponin extract(0.22 g/kg),phenolic acid extract(0.17 g/kg),and tannin extract(0.09 g/kg)for 14 days.After administration,albumin(ALB),creatine kinase(CK),lung dry-wet ratio(W/D),protein content in alveolar lavage fluid,heart index,lung index,the pathological tissue slices of heart and lung were observed and measured to comprehensively evaluate the Lishui(利水)effect of PHYTOLACCAE RADIX decoction and its components on Shangjiaoshuiyinneiting rat model.Results:Compared with the blank group,the model group showed reduced ALB content(P<0.01),protein content in alveolar lavage fluid,increased lung W/D,CK,heart index,and lung index(P<0.05 or P<0.01),and severely damaged heart and lung tissues,indicating that the Shangjiaoshuiyinneiting rat model was properly established.Compared with the model group,PHYTOLACCAE RADIX decoction(0.81 g/kg)increased serum ALB content(P<0.05)and reduced CK,protein content in alveolar lavage fluid,heart index,and lung W/D(P<0.05 or P<0.01).Polysaccharide extract(0.31 g/kg)significantly increased ALB content(P<0.05)and reduced CK,heart index,lung index,and lung W/D(P<0.05 or P<0.01).Saponin extract(0.22 g/kg)significantly increased ALB(P<0.05)and reduced CK(P<0.05).PHYTOLACCAE RADIX decoction(0.81 g/kg)and polysaccharide extract(0.31 g/kg)could markedly improve the pathological conditions of the heart and lung of model rats.Conclusion:P.acinosa decoction can improve heart and lung injury in Shangjiaoshuiyinneiting model rats.Polysaccharide and saponin extracts have therapeutic effects of different degrees on Shangjiaoshuiyinneiting model rats,in which polysaccharide extract shows more obvious therapeutic efficacy.
