摘要
目的:探究蒲公英根提取物对三阴性乳腺癌(Triple-negative breast cancer, TNBC)细胞增殖、迁移及侵袭能力的影响及其作用机制。方法:采用CCK-8法检测蒲公英根提取物对MDA-MB-231人乳腺癌细胞及4T1小鼠乳腺癌细胞增殖的抑制作用。通过划痕实验与Transwell侵袭实验检测蒲公英根提取物对两种细胞系迁移及侵袭能力的影响。利用RNA-seq测序分析蒲公英提取物对TNBC转录组的影响,并对Nanos C2HC型锌指蛋白1(Nanos C2HC-type zinc finger 1,Nanos1)的表达及患者预后进行分析。结果:与空白对照组相比,蒲公英根提取物组细胞增殖能力降低,迁移距离降低,侵袭数量下降(P<0.01);蒲公英根提取物组细胞Nanos1表达下调(P<0.01),且Nanos1的低表达与乳腺癌患者生存时间呈正相关(P<0.01)。结论:蒲公英根提取物通过下调Nanos1的表达抑制TNBC细胞增殖、迁移及侵袭能力。
Objective:To explore the effect of Taraxacum officinale root extract on the proliferation,migration and invasion of triple-negative breast cancer(TNBC)cells as well as its molecular mechanism.Methods:CCK-8 method was used to detect the inhibitory effect of T.officinale root extract on the proliferation of MDA-MB-231 human breast cancer cells and 4T1 mouse breast cancer cells.Wound healing assay and transwell assay were performed to verify the impact of T.officinale root extract on the migration and invasion of the two cell lines.RNA-seq sequencing(RNA-seq)was performed to observe the effect of T.officinale root extract on the transcriptome of TNBC cells,and the expression of Nanos C2HC-type zinc finger 1(Nanos1)and patient prognosis were analyzed.Results:Compared with the normal control group,the T.officinale root extract group presented decreased cell proliferation,migration and invasion(P<0.01)and down-regulated Nanos1 expression(P<0.01),and the low Nanos1 expression had positive correlation with the survival time of breast cancer patients(P<0.01).Conclusion:T.officinale root extract can inhibit the proliferation,migration and invasion of TNBC cells by down-regulating Nanos1 expression,which provides theoretical basis for T.officinale to be a candidate anti-cancer drug.
