LncRNA SPRY4-IT1调节血管平滑肌细胞在子宫螺旋动脉重铸中的作用

摘要 目的探讨LncRNA快速发育生长因子同源蛋白4内含子转录本1(SPRY4-IT1)在人早孕绒毛组织中的表达,以及SPRY4-IT1调节血管平滑肌细胞生物学行为在子宫螺旋动脉重铸中的作用。方法收集因孕早期胚胎停育行人工流产患者的绒毛组织及非计划妊娠行人工流产患者的绒毛组织,采用qRT-PCR检测两组胎盘组织中SPRY4-IT1的表达;利用原位杂交定位SPRY4-IT1在子宫螺旋动脉肌层的表达。结果与正常绒毛组织相比,SPRY4-IT1在胚胎停育绒毛组织中呈现高表达,差异有统计学意义(P<0.05);原位杂交结果显示SPRY4-IT1广泛表达于早孕蜕膜组织的螺旋动脉肌层。结论胚胎停育绒毛组织中SPRY4-IT1表达升高,其可能通过调节血管平滑肌细胞功能影响子宫螺旋动脉重铸过程。 Objective To investigate the expression of LncRNA growth factor homologous protein 4 intron Transcript 1(SPRY4-IT1)in human villi in early pregnancy and the role of SPRY4-IT1 in regulating the biological behavior of vascular smooth muscle cells in the recast of uterine spiral artery.Methods The chorionic tissues of patients with induced abortion due to termination of early pregnancy and those of patients with unplanned pregnancy were collected,and the expression of SPRY4-IT1 in placenta of the two groups was detected by qRT-PCR,and the expression of SPRY4-IT1 in the myometrium of uterine spiral artery was localized by in situ hybridization.Results Compared with normal chorionic villi,SPRY4-IT1 was highly expressed in aborted chorionic villi,and the results of in situ hybridization showed that SPRY4-IT1 was widely expressed in the muscular layer of spiral arteries in decidual tissues of early pregnancy.Conclusion The expression of SPRY4-IT1 in the villi of aborted embryos is increased,which may affect the recasting process of uterine spiral artery by regulating the function of vascular smooth muscle cells.
