叠鞘石斛组织培养及植株再生体系优化

摘要 以叠鞘石斛成熟种子为材料,使用IBA、6-BA和NAA共3种生长调节剂与香蕉泥和椰子汁有机物等组合,采用3因素3水平正交设计及单因素设计进行组织培养研究.结果显示,最佳叠鞘石斛原球茎培养基为MS+50 g/L椰子汁+30 g/L蔗糖+6 g/L琼脂,40 d增殖系数为6.75,分化率为84.38%;最佳叠鞘石斛壮苗生根培养基为1/2 MS+0.5 mg/L IBA+1.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L蔗糖+6 g/L琼脂.通过该技术,可以在较短时间获得大批量优质叠鞘石斛种苗,为大规模叠鞘石斛药材生产提供基础. Mature seeds of Dendrobium aurantiacum Rchb.f.var.denneanum(kerr)Z.H.Tsi were used as experimental materials,IBA,6-BA,NAA,mashed banana,and coconut juice were added in and three factors and three levels orthogonal design and single factor design were adopted.The study showed that the best corm culture medium was MS+50 g/L coconut juice+30 g/L sucrose+6 g/L agar,the proliferation coefficient was 6.75 and the differentiation efficiency was 84.38%at 40 days.The best culture medium for strong seedling and rooting was 1/2 MS+0.5 mg/L IBA+1.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L sucrose+6 g/L agar.It can be concluded that this tissue culture system can provide Dendrobium aurantiacum seedlings rapidly and also provide basis for the generous producation of this medicinal plants.
