基于网络药理学金荞麦治疗阿尔茨海默病机制探讨

摘要 目的:基于网络药理学,探索金荞麦治疗阿尔茨海默病(Alzheimer Disease,AD)的潜在物质基础、作用机制。方法:利用中药系统药理学数据库与分析平台(TCMSP),设定筛选主要药物成分条件为口服利用度≥30%、类药性≥0.18,筛选出金荞麦15种主要有效活性成分,登录GeneCards、OMIM等库检索AD的核心靶标。利用Cytoscape构建金荞麦治疗AD的“成分-靶标-疾病”网络,将共同关键核心靶点导入至String库,构建蛋白质-蛋白质相互作用网络图,对蛋白质-蛋白质相互作用进行可视化分析和拓扑参数分析。对靶点进行基因本体论(GO)、京都基因与基因组百科全书(KEGG)分析,探究金荞麦抗AD的物质基础及作用机制。结果:筛选出金荞麦15个主要有效活性成分,包括槲皮素、异鼠李素等主要有效成分;对用靶点包括白细胞介素-6、半胱氨酸蛋白酶3(Caspase 3,CASP3)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、血管内皮生长因子A(Vascular Endothelial Growth Factor A,VEGFA)、MYC原癌基因(MYC Proto-Oncogene,MYC)、雌激素受体1(Estrogen Receptor 1,ESR1)、丝裂原活化蛋白激酶3(Mitogen-Activated Protein Kinase 3,MAPK3)等;获取信号通路,如晚期糖基化终末产物-糖基化终末产物受体、肿瘤抑制蛋白p53、磷脂酰肌醇3激酶-蛋白激酶B等信号通路。结论:本次研究的结果可对金荞麦的药效进行全面认识和更深层次的研究提供实验与临床依据。 Objective:To explore the potential material basis and mechanism of Jin Qiaomai(Rhizoma Fagopyri Dibotryis)in the treatment of Alzheimer’s disease based on network pharmacology.Methods:TCMSP was used to screen the main active components of Jin Qiaomai.The conditions were as follows:oral availability≥30%,generic drug≥0.18.Fifteen main active components of Jin Qiaomai were screened.The core targets of AD were searched in GeneCards and OMIM.The component-target-disease network of Jin Qiaomai for AD treatment was constructed by using Cytoscape.The common key core targets were imported into the String library,and the PPI network diagram was constructed to analyze PPI visualization and topological parameters.Go and KEGG analysis were carried out to explore the material basis and mechanism of anti AD effect of Jin Qiaomai.Results:A total of 15 main active components of Jin Qiaomai were screened,including quercetin,isorhamnetin.The targets used included IL-6,CASP3,EGFR,VEGFA,MYC,ESR1,MAPK3,etc..The three main pathways were AGE/RAGE,P53 and PI3K/Akt.Conclusion:The results of this study can provide experimental and clinical basis for comprehensive understanding and deeper research on the efficacy of Jin Qiaomai.
