痛风平方剂对急性痛风性关节炎大鼠的治疗作用 被引量:1

The effect of the Tongfengping prescription on acute gouty arthritis in rats

摘要 目的:探索痛风平方剂对急性痛风性关节炎(Acute Gouty Arthritis,AGA)模型大鼠的治疗作用。方法:采用Coderre TJ的经典造模方法建立AGA大鼠模型。将48只雄性SD大鼠随机分为正常组、模型组、秋水仙碱组、痛风平高剂量组(0.12 g/100 g)、痛风平中剂量组(0.06 g/100 g)、痛风平低剂量组(0.03 g/100 g),每组大鼠8只。实验期间观察大鼠一般情况,给药干预结束后检测大鼠血清中尿酸、肌酐以及炎症因子(肿瘤坏死因子-α、白细胞介素-1β)浓度水平;测量计算大鼠关节肿胀度,记录步态变化,以及大鼠距小腿关节形态学观察。结果:痛风平方剂各剂量均可以减轻AGA大鼠模型关节肿胀率(P<0.001)和改善步态(P<0.05);痛风平方剂高剂量能降低血清中肌酐、肿瘤坏死因子-α和白细胞介素-1β水平(P<0.001)。结论:痛风平方剂对AGA大鼠有镇痛抗炎的作用,且能保护大鼠的肾功能。 Objective:To explore the therapeutic effects of the Tongfengping prescription(痛风平方)on acute gouty arthritis(AGA)in rats.Methods:The model of AGA in rats was established by Coderre TJ classic methods.Forty-eight male SpragueDawley rats were randomly divided into the normal group,the model group,the colchicine group(positive group),the Tongfengping high-dose group(0.12 g/100 g),the Tongfengping middle-dose group(0.06 g/100 g),the Tongfengping low-dose group(0.03 g/100 g),8 rats in each group.The general condition of the rats during the experiment was observed.After the end of the drug intervention,the concentrations of uric acid(UA),creatinine,and inflammatory factors(TNF-α,IL-1β)in the serum of the rats were detected.The degree of joint swelling,the gait score and morphological observation of talar joints of rats was observed.Results:Each dose of the Tongfengping prescription can reduce joint swelling rate(P<0.001)and ameliorate gait(P<0.05)in AGA rat models,high-dose of the Tongfengping prescription can reduce serum creatinine,TNF-αand IL-1βlevels(P<0.001).Conclusion:The Tongfengping prescription has analgesic and anti-inflammatory effects on AGA in rats,and can protect renal function in rats.
