血清LncRNA TINCR、miR-31表达水平与妊娠期糖尿病胰岛素抵抗的关系 被引量:1

Relationship between the expression levels of serum LncRNA TINCR and miR-31 and insulin resistance in gestational diabetes mellitus

摘要 目的探讨血清长链非编码RNA(LncRNA)TINCR、微小RNA-31(miR-31)表达水平与妊娠期糖尿病(GDM)胰岛素抵抗(IR)的关系。方法回顾性选取2019年5月至2021年12月首都医科大学大兴教学医院收治的98例经口服葡萄糖耐量试验(OGTT)判定为GDM的患者为GDM组,并纳入同期98名经OGTT确定为正常糖耐量(NGT)的孕妇为NGT组。比较两组临床资料、血清LncRNA TINCR、miR-31、白细胞介素(IL)-10、IL-17水平及稳态模型胰岛素抵抗指数(HOMA-IR);分析GDM患者血清LncRNA TINCR、miR-31表达水平与IL-10、IL-17、HOMA-IR,及血清LncRNA TINCR表达水平与miR-31的相关性;分析HOMA-IR的影响因素。结果GDM组患者空腹血糖、甘油三脂、空腹胰岛素(FINS)、miR-31、IL-17水平、HOMA-IR分别为(6.16±2.05)mmol/L、(2.97±0.99)mmol/L、(13.64±4.55)mIU/L、2.15±0.72、(118.72±39.57)ng/L、3.73±0.93,均高于NGT组[(4.73±1.58)mmol/L、(2.08±0.69)mmol/L、(6.80±2.27)mIU/L、1.01±0.34、(45.13±15.04)ng/L、1.43±0.36],血清LncRNA TINCR、IL-10水平分别为0.52±0.17、(154.38±51.46)ng/L,均低于NGT组[1.04±0.35、(195.62±65.21)ng/L],差异均有统计学意义(P<0.05)。GDM患者血清LncRNA TINCR表达水平与IL-10呈正相关(r=0.509,P<0.05),与IL-17、HOMA-IR呈负相关(r=-0.532、-0.497,P<0.05);而GDM患者血清miR-31表达水平与IL-10呈负相关(r=-0.483,P<0.05),与IL-17、HOMA-IR呈正相关(r=0.540、0.462,P<0.05)。GDM患者血清LncRNA TINCR表达水平与miR-31呈负相关(r=-0.587,P<0.05);LncRNA TINCR、miR-31、空腹血糖、FINS、IL-10、IL-17是HOMA-IR的影响因素(P<0.05)。结论GDM患者血清LncRNA TINCR表达水平较低,miR-31表达水平较高,均与HOMA-IR、IL-10、IL-17显著相关,为临床预防GDM IR提供新策略。 Objective To investigate the relationship between the expression levels of serum long non-coding RNA(LncRNA)TINCR and microRNA-31(miR-31)and insulin resistance(IR)in gestational diabetes mellitus(GDM).Methods From May 2019 to December 2021,98 patients who were diagnosed as GDM by oral glucose tolerance test(OGTT)in Daxing Teaching Hospital,Capital Medical University were collected as the GDM group,and 98 pregnant women who were determined as normal glucose tolerance(NGT)by OGTT in same course were included as NGT group.The clinical data,serum LncRNA TINCR,miR-31,interleukin(IL)-10,IL-17 levels and homeostatic model insulin resistance index(HOMA-IR)were compared between the two groups;the correlation of serum LncRNA TINCR and miR-31 expression levels with IL-10,IL-17,HOMA-IR,and the correlation of serum LncRNA TINCR expression level and miR-31 in GDM patients were analyzed;the influencing factors of HOMA-IR were analyzed.Results The fasting blood glucose,triglycerides,fasting insulin(FINS),miR-31,IL-17 levels,and HOMA-IR were(6.16±2.05)mmol/L,(2.97±0.99)mmol/L,(13.64±4.55)mIU/L,2.15±0.72,(118.72±39.57)ng/L,3.73±0.93 in the GDM group,which were higher than those in the NGT group[(4.73±1.58)mmol/L,(2.08±0.69)mmol/L,(6.80±2.27)mIU/L,1.01±0.34,and(45.13±15.04)ng/L,1.43±0.36],the levels of serum LncRNA TINCR,IL-10 were 0.52±0.17 and(154.38±51.46)ng/L,which were lower than those in the NGT group[1.04±0.35 and(195.62±65.21)ng/L],the differences were statistically significant(P<0.05).The expression level of serum LncRNA and TINCR in GDM patients was positively correlated with IL-10(r=0.509,P<0.05),but negatively correlated with IL-17,HOMA-IR(r=-0.532,-0.497,P<0.05);in GDM patients,the expression of miR-31 was negatively correlated with IL-10(r=-0.483,P<0.05),and positively correlated with IL-17,HOMA-IR(r=0.540,0.462,P<0.05).Serum LncRNA TINCR expression level in GDM patients was negatively correlated with miR-31(r=-0.587,P<0.05).LncRNA TINCR,miR-31,FBG,FINS,IL-10,and IL-17 were the influencing factors of HOMA-IR(P<0.05).Conclusion The expression level of serum LncRNA TINCR in GDM patients is low,and the expression level of miR-31 is high,they are obviously correlated with HOMA-IR,IL-10,and IL-17,and provide a new strategy for clinical prevention of GDM IR.
